Western blot analysis was performed using tissue lysates from human intestine, mouse intestine, rat intestine, hamster intestine, guinea pig intestine, and rabbit intestine probed with Intestinal FABP/Intestinal fatty acid-binding protein antibody (clone FABP2/6345). A distinct immunoreactive band is observed at the predicted molecular weight of FABP2 across all intestinal samples, consistent with conserved expression of Intestinal FABP in absorptive intestinal epithelium. Inset shows PBS substituted for the primary antibody as a secondary-only negative control. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95oC followed by cooling at RT for 20 minutes.

Analysis of Protein Array containing more than 19,000 full-length human proteins using recombinant GCG/Glucagon antibody (clone GCG/13090R). Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProtTM array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProtTM are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target. A MAb is considered to specific to its intended target, if the MAb has an S-score of at least 2.5. For example, if a MAb binds to protein X with a Z-score of 43 and to protein Y with a Z-score of 14, then the S-score for the binding of that MAb to protein X is equal to 29.

Additional closely spaced bands are observed in the same molecular weight range and are consistent with known GFAP isoforms and post-translationally modified species, which are well documented in the literature. The conserved multi-band pattern across species supports specific detection of GFAP-related protein forms rather than non-specific binding.
