BCL2L1 Antibody WB. Western blot analysis of human HeLa, Jurkat, HepG2, MCF7, and SH-SY5Y cell lysates using BCL2L1 Antibody demonstrates a prominent immunoreactive band at approximately 26-32 kDa, consistent with expression of BCL2L1/Bcl-XL. Bcl-XL is the major anti-apoptotic isoform encoded by the BCL2L1 gene and functions as a key regulator of mitochondrial apoptosis by inhibiting pro-apoptotic BCL2 family proteins and promoting cellular survival. A faint band near 50 kDa is observed in some cell lines and may represent an SDS-resistant Bcl-XL dimer or higher-order protein complex. This higher molecular weight species is substantially less abundant than the predominant monomeric form. Antibody incubation was performed at 1:1000 dilution for 1 hour at room temperature. Predicted molecular weight of Bcl-XL: approximately 26-30 kDa.

Bcl-XL Antibody FACS. Flow cytometry analysis of fixed and permeabilized human Jurkat cells using Bcl-XL Antibody / Anti-Apoptotic Protein Antibody demonstrates a pronounced rightward shift of the antibody-stained population (blue) relative to both the isotype control (green) and unstained cells (red), consistent with intracellular expression of Bcl-XL. Bcl-XL, encoded by the BCL2L1 gene, is a key anti-apoptotic member of the BCL2 protein family that promotes cellular survival by inhibiting mitochondrial apoptosis pathways and suppressing activation of pro-apoptotic signaling proteins. The observed staining pattern supports detection of Bcl-XL expression in Jurkat cells by flow cytometry. Cells were fixed, permeabilized, and stained with Bcl-XL antibody at 1 ug/million cells following blocking with goat sera. Red = cells alone, Green = isotype control, Blue = Bcl-XL antibody.

Immunofluorescent staining of PGAM5 using anti-PGAM5 antibody (green). PGAM5 was detected in an immunocytochemical section of human A549 cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 5 ug/ml rabbit anti-PGAM5 antibody overnight at 4oC. DyLight 488 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1:500 dilution and incubated for 30 minutes at 37oC. The section was counterstained with DAPI nuclear stain (blue). Visualize using a fluorescence microscope and filter sets appropriate for the label used.
