Bcl-X Antibody Hodgkin's Lymphoma IHC. Immunohistochemistry staining of Hodgkin's lymphoma tissue using Bcl-X Antibody (clone BX006) demonstrates cytoplasmic and membranous HRP-DAB brown staining within neoplastic cells. The staining pattern is consistent with expression of Bcl-X family proteins, including the anti-apoptotic isoform Bcl-XL, which functions to promote cellular survival through inhibition of mitochondrial apoptosis pathways. Bcl-X proteins are important regulators of programmed cell death and have been extensively studied in hematologic malignancies due to their roles in apoptosis resistance, tumor cell persistence, and therapeutic response. The observed cytoplasmic and membrane-associated staining supports expression of Bcl-X in lymphoma-associated cellular populations.
Bcl-XL Antibody WB. Western blot analysis of human Jurkat cell lysate using Bcl-XL Antibody / Anti-Apoptotic Protein Antibody, clone BX006, demonstrates a distinct immunoreactive band at approximately 26-30 kDa, consistent with expression of Bcl-XL, the major anti-apoptotic isoform encoded by the BCL2L1 gene. Bcl-XL functions as a key regulator of mitochondrial apoptosis by inhibiting pro-apoptotic BCL2 family proteins and promoting cellular survival under conditions of physiological and cellular stress. The detected band is consistent with the expected molecular weight of Bcl-XL and supports specific recognition of endogenous BCL2L1 protein in Jurkat cells. Predicted molecular weight: 26-30 kDa.

Description

This Bcl-X Antibody, clone BX006, recognizes a protein of 27kDa, identified as Bcl-X. The mAb shows no cross-reaction with Bcl2 alpha or Bax. Bcl-X has two isoforms, XL (long), a 241 amino acid protein which suppresses cell death, and XS (short), a 178 amino acid protein lacking a 63 amino acid domain which functions as a dominant inhibitor of Bcl2.

Application Notes

The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the Bcl-X antibody to be titered up or down for optimal performance.

1. Staining of formalin-fixed tissues requires boiling tissue sections in 1mM EDTA buffer, pH 7.5-8.5, for 10-20 min followed by cooling at RT for 20 minutes.
2. This antibody reacts with both BclXS and BclXL.
3. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

Immunogen

Amino acids QSNRELVVDFLS (3-14) of the human protien were used as the immunogen for this Bcl-X antibody (2).

Storage

Store the Bcl-X antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
