Rabbit Monoclonal Antibody / Antibodies

The rabbit monoclonal antibody is a relatively new addition to the research antibody portfolio of mouse/rat monoclonals, and rabbit/donkey polyclonals. Some of the advantages of a rabbit monoclonal antibody are:

  • increased sensitivity to mouse antigens
  • unlike with mouse mAbs, secondary antibody will not cross-react with mouse immunoglobulin in test samples
  • a general lower background
  • a general increase in affinity allowing for less rabbit monoclonal antibody to be used per test

    Immunohistochemical staining of Neurokinin 1 Receptor using anti-NKR1 antibody. Neurokinin 1 Receptor was detected in a paraffin-embedded section of mouse colon tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1:50 rabbit anti-NKR1 antibody overnight at 4oC. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37oC. The tissue section was developed using an HRP secondary and DAB substrate.

    Western blot analysis of NOLC1 using anti-NOLC1 antibody. Lane 1: human Hela whole cell lysates, Lane 2: human CACO-2 whole cell lysates, Lane 3: human whole cell lysates, Lane 4: human MCF-7 whole cell lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-NOLC1 antibody at 1:500 overnight at 4oC, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:500 for 1.5 hour at RT. The signal was developed using enhanced chemiluminescent. NOLC1 western blot across human cell lines shows a predominant band at ~130 kDa. Although the predicted mass is ~74 kDa, NOLC1/Nopp140 is highly acidic and heavily phosphorylated, causing it to migrate markedly above its theoretical size on SDS-PAGE.
