Cytokeratin 17 Antibody / KRT17 Epithelial Marker detects keratin 17 (KRT17), also known as cytokeratin 17, CK17 or K17, a type I intermediate filament protein expressed in selected epithelial cell populations. Keratin intermediate filaments are major components of the epithelial cytoskeleton, where they contribute to mechanical stability, cellular architecture and resistance to physical stress. KRT17 has a characteristic distribution in basal epithelial compartments and specialized epithelial structures, making it an important marker for studying epithelial identity, differentiation and tissue organization.

KRT17 forms intermediate filament networks through association with type II keratin partners, particularly keratin 5 and keratin 6. These cytoplasmic filament systems provide structural support while participating in dynamic changes associated with epithelial growth and differentiation. In normal tissues, Cytokeratin 17 is found in selected basal and myoepithelial cell populations and in structures such as hair follicles and other appendageal epithelia. Its relatively restricted expression compared with broadly distributed epithelial keratins makes a Cytokeratin 17 Antibody useful for identifying KRT17-positive populations and investigating epithelial lineage.

Cytokeratin 17 is also responsive to changes in epithelial cell state. KRT17 expression can increase during wound healing, tissue regeneration and other conditions involving epithelial activation or hyperproliferation. Beyond its structural contribution to intermediate filaments, KRT17 has been linked to cellular processes associated with growth, stress responses and inflammatory signaling. These characteristics have expanded interest in the KRT17 epithelial marker as a research target for understanding how epithelial cells adapt during tissue injury, remodeling and disease.

Altered KRT17 expression is particularly relevant to cancer biology. Cytokeratin 17 has been investigated in a variety of epithelial malignancies, including squamous cell carcinomas and tumors of the breast, lung, pancreas and other tissues. Its expression can help define basal or squamous differentiation patterns within heterogeneous tumors and is frequently studied alongside other cytokeratins and epithelial markers. KRT17 expression has also been associated with aggressive tumor phenotypes in several cancer types, supporting continued investigation of its relationship to tumor cell proliferation, differentiation, invasion and disease progression.

Clone E3 is a well-established mouse monoclonal antibody directed against Cytokeratin 17. The antibody was generated against a cytoskeletal preparation from rat colon epithelium and recognizes the KRT17 intermediate filament protein. Application Notes

The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the Cytokeratin 17 Antibody E3 / KRT17 Epithelial Marker to be titered up or down for optimal performance.

1. Staining of formalin-fixed tissues requires boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 10-20 min followed by cooling at RT for 20 minutes.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

Immunogen

The cytoskeletal fraction of rat colon epithelium was used as the immunogen for this Cytokeratin 17 antibody.

Storage

Store the Cytokeratin 17 antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
