Description

PSA Antibody A67-B/E3 recognizes prostate specific antigen (PSA), a secreted serine protease encoded by the KLK3 gene. PSA belongs to the kallikrein related peptidase family and is produced predominantly by luminal epithelial cells of the prostate gland. The protein is synthesized as a precursor and undergoes proteolytic processing to generate the mature enzyme that is secreted into seminal fluid.

PSA functions primarily in liquefaction of the seminal coagulum through proteolytic cleavage of semenogelins and other seminal proteins. The mature protein is glycosylated and can occur in both free and protein-bound forms. In circulation, PSA can form complexes with endogenous protease inhibitors, particularly alpha 1-antichymotrypsin, contributing to the different molecular forms of PSA studied in prostate biology.

KLK3 expression is strongly associated with differentiated secretory prostate epithelium and is regulated by androgen receptor signaling. PSA expression is commonly retained in prostate adenocarcinoma and prostate-derived tumors, making it useful for research into prostate epithelial differentiation, tumor lineage, androgen-regulated biology, and molecular changes associated with prostate cancer.

Clone A67-B/E3 is a mouse monoclonal antibody generated against prostate specific antigen purified from human sperm plasma. This established clone provides a defined reagent for investigating PSA/KLK3 protein expression and has also been used as a complementary antibody in PSA assay research. As a Prostate Specific Antigen MAb, clone A67-B/E3 supports studies examining prostate-associated protein expression and tumor biology. NSJ Bioreagents supplies A67-B/E3 for research use only.

Application Notes

The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the PSA Antibody A67-B/E3 / Prostate Specific Antigen MAb to be titered up or down for optimal performance.

1. Staining of formalin-fixed tissues requires boiling tissue sections in 1mM EDTA Buffer, pH 7.5-8.5, for 10-20 min followed by cooling at RT for 20 minutes.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

Prostate Specific Antigen from human sperm plasma was used as the antigen for this PSA Antibody A67-B/E3.

Storage

Store the PSA Antibody A67-B/E3 at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
