PSA Antibody Rabbit Monoclonal KLK3/2871R IHC staining of human prostate carcinoma. Immunohistochemistry analysis of FFPE human prostate carcinoma tissue using PSA Antibody Rabbit Monoclonal KLK3/2871R shows HRP-DAB brown cytoplasmic staining in malignant prostate epithelial cells forming glandular tumor structures, consistent with Prostate specific antigen (KLK3) expression in prostate-derived carcinoma cells, while surrounding stromal cells remain largely negative. HIER: tissue sections were boiled in 10mM Tris with 1mM EDTA, pH 9 for 10-20 minutes and allowed to cool before testing.

Description

Prostate specific antigen (KLK3) is a secreted serine protease encoded by the KLK3 gene and produced predominantly by luminal epithelial cells of the prostate gland. The protein is widely known as Prostate specific antigen or PSA and belongs to the kallikrein related peptidase family of serine proteases. PSA Antibody Rabbit Monoclonal KLK3/2871R recognizes the KLK3 protein and enables detection of PSA expression in studies focused on prostate epithelial biology and prostate cancer.

PSA is synthesized as a precursor enzyme that contains a signal peptide and propeptide sequence that are removed during secretion and enzymatic activation. The mature enzyme is released into the lumen of prostatic glands where it functions as a protease in seminal plasma. PSA cleaves semenogelin proteins and related substrates, promoting liquefaction of the seminal coagulum after ejaculation and contributing to normal reproductive physiology.

In normal prostate tissue, KLK3 expression is largely restricted to luminal epithelial cells lining prostatic acini and ducts. The protein is typically localized to the cytoplasm and luminal secretions of glandular epithelial cells, reflecting active synthesis and secretion of PSA by prostate epithelium. This highly restricted tissue distribution has made PSA one of the most widely used markers for identifying prostate epithelial differentiation in biological and cancer research studies.

PSA expression is commonly retained in prostate adenocarcinoma and in metastatic tumors derived from prostate tissue. Detection of Prostate specific antigen therefore provides valuable biological information when investigating prostate tumor differentiation, tumor origin, and the cellular lineage of prostate-derived malignancies. Analysis of PSA expression patterns can help researchers examine prostate tumor progression and the biological characteristics of prostate cancer cells.

Application Notes

1. The optimal dilution of the PSA Antibody Rabbit Monoclonal KLK3/2871R for each application should be determined by the researcher.

2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

3. This MAb makes an excellent pair with mAb A67-B/E3 for PSA tests.
