Band 3 Antibody Human Placenta IHC. Immunohistochemical staining of FFPE human placenta tissue using Band 3 Antibody / Anion Exchange Protein 1 Antibody demonstrates strong membranous and cytoplasmic HRP-DAB brown staining within placental trophoblast cell populations. Band 3 (AE1/SLC4A1) is a chloride/bicarbonate exchanger that mediates transmembrane ion transport and contributes to maintenance of acid-base balance and cellular ionic homeostasis. In addition to its well-established role in erythrocyte membrane physiology, Band 3 family proteins participate in transport processes that support cellular metabolism and physiologic homeostasis in specialized epithelial tissues. The observed placental staining pattern is consistent with expression of anion transport machinery involved in regulation of bicarbonate and chloride exchange. HIER: boil paraffin sections in pH 6, 10 mM citrate buffer for 20 minutes and allow to cool prior to staining.

As one of the most abundant membrane proteins in red blood cells, SLC4A1 contributes to membrane organization, structural stability, and physiologic ion transport. HIER: boil tissue sections in pH 8 EDTA for 20 minutes and allow to cool before testing.

Immunohistochemical staining of paraffin-embedded zebrafish gill tissue using Zebrafish SLC4A1 Antibody / SLC4A1A Antibody demonstrates strong HRP-DAB brown staining within gill filament-associated cells and erythrocyte-rich structures. The staining pattern is consistent with expression of SLC4A1A, a chloride/bicarbonate exchanger that contributes to ion transport, acid-base regulation, and erythrocyte physiology. As the zebrafish ortholog of human SLC4A1 (Band 3/AE1), SLC4A1A plays an important role in maintaining cellular ionic balance and supporting carbon dioxide transport. Expression within gill-associated tissues is consistent with the involvement of anion transport proteins in respiratory physiology, gas exchange, and regulation of the aquatic ionic environment. Heat-mediated antigen retrieval was performed in EDTA buffer prior to overnight primary antibody incubation and HRP-DAB visualization.
