Rabbit Monoclonal Antibody / Antibodies

The rabbit monoclonal antibody is a relatively new addition to the research antibody portfolio of mouse/rat monoclonals, and rabbit/donkey polyclonals. Some of the advantages of a rabbit monoclonal antibody are:

  • increased sensitivity to mouse antigens
  • unlike with mouse mAbs, secondary antibody will not cross-react with mouse immunoglobulin in test samples
  • a general lower background
  • a general increase in affinity allowing for less rabbit monoclonal antibody to be used per test

    Western blot testing of human samples using the PALLD antibody at 1:3000 dilution for 1 hour at room temperature. PALLD antibody detects a band migrating at ~125-135 kDa in the indicated cell lysates, consistent with the 140-kDa Palladin isoform. Minor gel-to-gel variation reflects phosphorylation-dependent mobility and cytoskeletal protein behavior. Lower fragments (when present) are attributed to regulated proteolysis.

    Immunohistochemical staining of CD163L1 using anti-CD163L1 antibody. CD163L1 was detected in a paraffin-embedded section of human appendix tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with a dilution of 1:50 rabbit anti-CD163L1 antibody overnight at 4oC. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37oC. The tissue section was developed using an HRP secondary and DAB substrate.

    Western blot analysis of Histone H3 (acetyl K9) using anti-Histone H3 (acetyl K9) antibody. Electrophoresis was performed on a 12% SDS-PAGE gel at 80V (Stacking gel) / 120V (Resolving gel) for 2 hours. Lane 1: human 293T whole cell lysates, Lane 2: human U20S whole cell lysates, Lane 3: human T98G whole cell lysates, Lane 4: human whole cell lysates, Lane 5: human K562 whole cell lysates, Lane 6: human SH-SY5Y whole cell lysates, Lane 7: human U251 whole cell lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Histone H3 (acetyl K9) antibody at 1:500 overnight at 4oC, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal was developed using an ECL Plus Western Blotting Substrate. A specific band was detected for Histone H3 (acetyl K9) at approximately 15 kDa. The expected molecular weight of Histone H3 (acetyl K9) is at 15 kDa.
