Rabbit Monoclonal Antibody / Antibodies

The rabbit monoclonal antibody is a relatively new addition to the research antibody portfolio of mouse/rat monoclonals, and rabbit/donkey polyclonals. Some of the advantages of a rabbit monoclonal antibody are:

  • increased sensitivity to mouse antigens
  • unlike with mouse mAbs, secondary antibody will not cross-react with mouse immunoglobulin in test samples
  • a general lower background
  • a general increase in affinity allowing for less rabbit monoclonal antibody to be used per test

    Immunohistochemical staining of Ki67/MKI67 using anti-Ki-67 antibody. Ki67/MKI67 was detected in a paraffin-embedded section of mouse spleen tissue. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 1:50 rabbit anti-Ki-67 antibody overnight at 4oC. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37oC. The tissue section was developed using an HRP secondary and DAB substrate.

    Immunohistochemical staining using SLC25A12 antibody on paraffin-embedded human colon cancer tissue sections. Heat-induced epitope retrieval was performed using EDTA buffer (pH 8.0) prior to antibody incubation. Sections were blocked with 10% goat serum and incubated with SLC25A12 antibody overnight at 4C. Detection was carried out using an HRP-based secondary antibody with DAB as the chromogen. Staining shows cytoplasmic immunoreactivity in tumor cells. Nuclei were counterstained with hematoxylin.

    Western blot analysis of BDKRB2 using anti-BDKRB2 antibody. Lane 1: human Hela whole cell lysates, Lane 2: human 293T whole cell lysates, Lane 3: human MCF-7 whole cell lysates, Lane 4: human K562 whole cell lysates, Lane 5: mouse lung tissue lysates, Lane 6: mouse kidney tissue lysates. After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-BDKRB2 antibody at 1:500 overnight at 4oC, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:500 for 1.5 hour at RT. The signal was developed using enhanced chemiluminescent. A specific band was detected for BDKRB2 at approximately 78 kDa. The expected molecular weight of BDKRB2 is ~78 kDa.
