Analysis of Protein Array containing >19,000 full-length human proteins using EpCAM Mouse Monoclonal Antibody (EGP40/1372). Z- and S- Score: The Z-score represents the strength of a signal that a monoclonal antibody (MAb) (in combination with a fluorescently-tagged anti-IgG secondary antibody) produces when binding to a particular protein on the HuProtTM array. Z-scores are described in units of standard deviations (SD's) above the mean value of all signals generated on that array. If targets on HuProtTM are arranged in descending order of the Z-score, the S-score is the difference (also in units of SD's) between the Z-score. S-score therefore represents the relative target specificity of a MAb to its intended target. A MAb is considered to specific to its intended target, if the MAb has an S-score of at least 2.5. For example, if a MAb binds to protein X with a Z-score of 43 and to protein Y with a Z-score of 14, then the S-score for the binding of that MAb to protein X is equal to 29.

Description

This antibody recognizes an oncofetal glycoprotein with a single chain of 70kDa, which is identified as Alpha fetoprotein (AFP). The AFP antibody is highly specific and shows no cross-reaction with other oncofetal antigens or serum albumin. Alpha fetoprotein is normally synthesized in the liver, intestinal tract, and yolk sac of the fetus. AFP antibody has been shown to be useful in detecting hepatocellular carcinomas (HCC) and germ cell neoplasms, especially yolk sac tumors.

This antibody can be compared with our AFP Antibody cocktail (clones C2 + C3 + MBS-12) for robust detection of alpha fetoprotein across multiple assay formats in tumor marker studies.

Application Notes

The concentration stated for each application is a general starting point. Variations in protocols, secondaries and substrates may require the AFP antibody to be titered up or down for optimal performance.

1. No special pretreatment is required for the immunohistochemical staining of formalin-fixed, paraffin-embedded tissues.
2. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.

Immunogen

Alpha fetoprotein purified from serum of a hepatoma patient was used as the immunogen for this AFP antibody.

Storage

Store the AFP antibody at 2-8oC.

Alternate Names

Alpha fetoglobulin; FETA; HPAFP, Alpha fetoprotein, AFP antibody
