CD38 Antibody / Plasma Cell and Immune Activation Marker Antibody

CD38 Antibody Overview

CD38 (CD38) is a type II transmembrane glycoprotein with ectoenzyme activity that regulates NAD metabolism and calcium signaling, playing a key role in immune cell activation, proliferation, and differentiation. It is broadly expressed on plasma cells, activated T and B lymphocytes, natural killer cells, and select myeloid populations, making it an important marker for immune system function and regulation.

A CD38 Antibody is widely used to detect CD38 expression across immune cell populations in both normal and disease contexts. CD38 antibody labeling is especially prominent in plasma cells, where strong surface and cytoplasmic expression supports identification of antibody-producing cells within lymphoid tissues and inflammatory environments. In addition to plasma cells, CD38 is expressed on activated lymphocytes, allowing it to serve as a marker of immune activation and cellular differentiation.

In normal tissues, CD38 expression is most abundant in lymphoid organs such as tonsil, spleen, and lymph node, where plasma cells and activated immune cells are present. In disease settings, CD38 is a key marker in hematologic malignancies including multiple myeloma and certain lymphomas, as well as in studies of immune infiltration within solid tumors. Its expression in the tumor microenvironment has gained increasing attention, particularly in relation to immune regulation and metabolic signaling pathways.

Because CD38 functions both as a surface marker and as an enzyme involved in NAD metabolism, it provides insight into both cellular identity and functional state. This dual role makes CD38 antibodies valuable tools in immunology, oncology, and cell biology research.

Applications of CD38 Antibodies

CD38 antibodies are used across multiple research applications depending on experimental needs:

Immunohistochemistry (IHC)

Used to visualize plasma cells and immune cell infiltration within tissue sections, highlighting cellular distribution and tissue architecture.

Flow Cytometry (FACS)

Enables identification and isolation of CD38-positive immune cell populations, including activated lymphocytes and plasma cells.

Western Blot (WB)

Supports detection of CD38 protein expression in cell lysates for analysis of protein levels and cellular regulation.

Immunofluorescence (IF)

Allows spatial visualization of CD38 expression and co-localization with other immune markers in cells and tissues.

CD38 as a Plasma Cell and Immune Marker

CD38 is widely recognized as a defining marker of plasma cell differentiation and immune activation. Its high expression in plasma cells makes it essential for identifying antibody-secreting cells, while its presence on activated lymphocytes supports its role in monitoring immune responses.

In addition to its use as a lineage marker, CD38 participates directly in enzymatic processes that regulate intracellular signaling through NAD metabolism and calcium mobilization. This functional role links CD38 expression to broader biological processes including immune regulation, inflammation, and tumor progression.

Choosing the Right CD Antibody

Selection of a CD antibody should be based on the intended research application and experimental system. Different antibody formats and clones are optimized for specific uses, including tissue-based detection, cell surface analysis, and protein expression studies.

Researchers may also consider factors such as clone performance, validation data, and specificity when selecting the most appropriate CD38 antibody for their work.
