Cytokeratin Antibody - Epithelial Lineage and Carcinoma Marker

Overview

Cytokeratins are a family of intermediate filament proteins expressed in epithelial cells and are widely used as markers of epithelial lineage. Cytokeratin antibody reagents enable detection of these structural proteins across a broad range of tissues, supporting identification of epithelial cells in both normal and disease contexts.

A Cytokeratin antibody is commonly used to distinguish epithelial-derived cells from non-epithelial populations, particularly in complex tissue environments. Because cytokeratin expression is retained in most carcinomas, these antibodies are essential tools for identifying epithelial tumors and evaluating tissue origin.

Cytokeratin Expression in Epithelial Cells

Cytokeratins are expressed in a tissue-specific and differentiation-dependent manner across epithelial cell types. Different epithelial tissues exhibit distinct cytokeratin profiles, reflecting their structural and functional specialization.

Simple epithelia typically express keratins such as KRT8 and KRT18, while stratified epithelia express keratins such as KRT5 and KRT14 in basal layers and KRT1 and KRT10 in differentiated layers. Glandular and transitional epithelia display additional keratin expression patterns that can be used to define tissue identity.

Because of this diversity, cytokeratin antibodies provide a reliable means of mapping epithelial organization and identifying cell populations within heterogeneous tissues.

Cytokeratin Antibodies in Cancer Identification

Cytokeratin expression is a defining feature of epithelial-derived tumors. Carcinomas retain cytokeratin proteins, while most mesenchymal, hematopoietic, and neural tumors do not, making cytokeratin antibodies essential for tumor classification.

Cytokeratin antibody staining is widely used to confirm epithelial origin in tumor samples and to distinguish carcinomas from other malignancies. In metastatic settings, cytokeratin detection helps identify the primary tissue source based on expression patterns.

Cytokeratin vs Individual Keratin Targets

Broad cytokeratin antibodies are commonly used for general epithelial identification, particularly when confirming epithelial origin across diverse tissue types. These reagents detect multiple keratin proteins and provide a high-level view of epithelial presence.

In contrast, individual keratin targets provide more detailed insight into epithelial subtype and differentiation. Selection of a specific keratin antibody depends on the biological context and the level of specificity required.

Applications of Cytokeratin Antibodies

Cytokeratin antibodies are used across multiple experimental approaches to study epithelial cells and tissue organization.

Immunohistochemistry

Cytokeratin antibodies are widely used in tissue sections to identify epithelial cells and visualize tissue architecture, particularly in carcinoma detection.

Western Blot

Detection of cytokeratin proteins allows confirmation of expression and comparison across samples.

Immunofluorescence

Cytokeratin antibodies enable visualization of intracellular filament organization and epithelial morphology.

Flow Cytometry

Cytokeratin antibodies can be used to identify epithelial-derived cells in mixed populations following appropriate preparation.

Relationship to Keratin (KRT) Antibody Targets

While cytokeratin antibodies are often used broadly to detect epithelial cells, keratin proteins are encoded by individual KRT genes that define specific epithelial subtypes and differentiation states.

Antibodies targeting individual keratin proteins, such as KRT1 antibody, KRT5 antibody, KRT7 antibody, and KRT19 antibody, provide more detailed insight into epithelial biology and are used when precise characterization of tissue type or differentiation status is required.

Summary

Cytokeratin antibody reagents provide a reliable approach for identifying epithelial cells, characterizing tissue organization, and distinguishing epithelial tumors. By detecting keratin proteins across a range of epithelial contexts, these antibodies support research into tissue structure, differentiation, and disease.
