ZBTB16 Antibody / Zinc Finger and BTB Domain Containing 16 Antibody recognizes ZBTB16, a sequence-specific transcription factor belonging to the BTB/POZ zinc finger family of transcriptional regulators. ZBTB16 functions primarily as a transcriptional repressor by recruiting chromatin remodeling complexes and histone deacetylases to target genes, thereby regulating gene expression programs that control cellular proliferation, differentiation, and lineage commitment. The protein contains an amino-terminal BTB/POZ domain that mediates protein-protein interactions and multiple carboxyl-terminal C2H2 zinc finger motifs responsible for sequence-specific DNA binding. Through these structural domains, ZBTB16 serves as an important regulator of development, stem cell maintenance, and tissue homeostasis.

ZBTB16 integrates developmental and environmental signals by controlling genes involved in hematopoiesis, skeletal development, spermatogenesis, and immune cell differentiation. It plays particularly important roles in hematopoietic stem cells, spermatogonial stem cells, natural killer T cells, innate lymphoid cells, and multiple lymphocyte populations. By repressing or activating specific transcriptional programs through chromatin modification, ZBTB16 helps maintain cellular identity while regulating the balance between self-renewal and differentiation. The protein also contributes to signaling pathways downstream of retinoic acid, steroid hormones, and cytokines, allowing extracellular cues to influence long-term patterns of gene expression.

Although ZBTB16 is the official gene symbol, the protein is widely recognized in the literature by its historical name, Promyelocytic Leukemia Zinc Finger (PLZF), which originated from its discovery as a fusion partner in a chromosomal translocation associated with acute promyelocytic leukemia. Beyond leukemia, altered ZBTB16 expression has been implicated in lymphoma, prostate cancer, melanoma, and several other malignancies, as well as developmental disorders and inflammatory diseases. The protein is predominantly localized within the nucleus, where it associates with chromatin to regulate transcription and epigenetic programs that influence cell fate decisions.
