STAM2 Antibody Human Spleen WB. Western blot testing of human spleen lysate using STAM2 Antibody / Hbp Antibody at 1 ug/ml. A specific band is detected at approximately 70 kDa, consistent with the routinely observed migration of STAM2 despite its predicted molecular weight of approximately 58 kDa. The higher apparent molecular weight is attributed to phosphorylation-dependent mobility shifts and the presence of multiple STAM2 isoforms. STAM2 Antibody / Hbp Antibody recognizes Signal Transducing Adaptor Molecule 2, a cytoplasmic adaptor protein historically known as Hbp (Hrs-binding protein) because of its direct interaction with Hrs (HGS). Hbp is a core component of the ESCRT-0 complex, where it cooperates with Hrs to recognize ubiquitinated membrane proteins and direct them into the endosomal sorting pathway. Through this activity, Hbp helps determine whether activated cell-surface receptors are recycled back to the plasma membrane or targeted for lysosomal degradation, making it an important regulator of receptor signaling and intracellular protein turnover.

Hbp contains an SH3 domain and multiple protein interaction motifs that allow it to function as a scaffold for signaling and membrane trafficking proteins. Together with Hrs, Hbp recruits ubiquitinated cargo and additional ESCRT machinery required for multivesicular body formation. This process is essential for the downregulation of receptor tyrosine kinases, cytokine receptors, and other signaling molecules following activation. By controlling receptor trafficking, Hbp influences signaling pathways involved in cell growth, differentiation, immune responses, and survival. Dysregulation of Hbp-mediated endosomal sorting has been linked to abnormal receptor signaling, impaired protein homeostasis, and cancer progression.

Because Hbp functions at the intersection of signal transduction and vesicle trafficking, it is widely studied in cell biology, oncology, neuroscience, and immunology. The protein localizes predominantly to early endosomes, where it displays a characteristic punctate cytoplasmic distribution that frequently concentrates in the perinuclear region. Studies of Hbp often focus on receptor internalization, endosomal maturation, ubiquitin-dependent cargo recognition, ESCRT complex assembly, and lysosomal targeting. Hbp also participates in the cellular response to growth factors and cytokines by regulating the duration and intensity of downstream signaling events.
