iRhom2 Antibody / Inactive Rhomboid Protein 2 Antibody detects iRhom2, a catalytically inactive member of the rhomboid protein family encoded by the RHBDF2 gene. Unlike proteolytically active rhomboid proteins, iRhom2 lacks functional protease activity and instead participates in membrane protein trafficking and signaling. A major function of iRhom2 is the regulation of ADAM17, a membrane-associated metalloprotease involved in the shedding of cytokines, growth factors and other cell-surface proteins.
Inactive rhomboid protein 2 contributes to the maturation, trafficking and regulation of ADAM17 as it moves through the secretory pathway toward the cell surface. Through its effects on ADAM17, iRhom2 can influence the release of membrane-bound substrates including tumor necrosis factor (TNF) and epidermal growth factor receptor (EGFR) ligands. These functions connect RHBDF2 with pathways controlling inflammation, cell proliferation, survival and tissue homeostasis.
iRhom2 is particularly relevant to immune and inflammatory signaling. Regulation of ADAM17-mediated TNF shedding provides a mechanism through which RHBDF2 can influence inflammatory responses, while release of EGFR ligands connects iRhom2 activity with growth factor signaling. The iRhom2-ADAM17 axis is therefore studied in inflammation, immune cell function, tissue repair and cancer biology.
Genetic alterations affecting RHBDF2 have been associated with tylosis with esophageal cancer, an inherited condition characterized by palmoplantar keratoderma and increased susceptibility to esophageal squamous cell carcinoma. Altered iRhom2 function is also investigated in diseases involving abnormal inflammatory or growth factor signaling. These relationships make inactive rhomboid protein 2 an important target for studies connecting membrane trafficking with cytokine and receptor signaling.

E. coli-derived recombinant human protein (amino acids H145-D529) was used as the immunogen for the RHBDF2 / iRhom2 antibody.
After reconstitution, the RHBDF2 / iRhom2 antibody can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.
