Interferon gamma Antibody rIFNG/4467 Detection in A549 Human Lung Cancer Cells WB. Western blot analysis of human A549 lung carcinoma cell lysate using Interferon gamma Antibody, clone rIFNG/4467. A prominent immunoreactive band is detected within the expected 20-25 kDa range for human interferon gamma. The discrete target signal and limited additional background demonstrate detection of Type II Interferon in A549 cells under the tested conditions.

SDS-PAGE analysis of purified, BSA-free Interferon gamma Antibody, clone rIFNG/4467, with 2 ug loaded under non-reducing (NR) and reducing (R) conditions. Under non-reducing conditions, the intact antibody migrates predominantly near 150 kDa. Under reducing conditions, bands corresponding to the antibody heavy and light chains are visible at approximately 50 kDa and 25 kDa, respectively. The observed banding pattern supports the integrity and purity of the recombinant monoclonal Type II Interferon Antibody preparation.

HIER was performed by boiling tissue sections in pH 9 10 mM Tris with 1 mM EDTA for 20 min followed by cooling before staining. Interferon gamma Antibody rIFNG/4467 / Type II Interferon Antibody recognizes interferon gamma (IFNG), a cytokine that serves as the sole member of the type II interferon class. IFNG is a central regulator of innate and adaptive immunity and is produced primarily by activated T lymphocytes and natural killer cells. Unlike the type I interferons, which include multiple interferon alpha and beta proteins, interferon gamma signals through a distinct receptor complex and activates cellular programs involved in immune defense, inflammation, and antigen recognition.

Interferon gamma binds the heterodimeric IFNGR1/IFNGR2 receptor complex, initiating JAK-STAT signaling dominated by activation of STAT1. This pathway promotes transcription of genes involved in antigen processing and presentation, antimicrobial defense, chemokine production, and immune cell communication. IFNG can enhance major histocompatibility complex expression and is particularly important in macrophage activation and cell-mediated immune responses. Through these functions, the Type II Interferon has an important role in coordinating immune activity against intracellular pathogens and other cellular challenges.

Dysregulated IFNG signaling has been investigated in chronic inflammatory and autoimmune disorders, infectious disease, and cancer. Within the tumor microenvironment, interferon gamma can contribute to antitumor immunity by enhancing antigen presentation and immune recognition, while prolonged signaling may also induce regulatory pathways that influence immune escape. These context-dependent effects have made the IFNG pathway an important area of research in tumor immunology, immunotherapy, inflammatory signaling, and host defense.
