Integrin Associated Protein Antibody rCD47/6365 in Human Prostate Carcinoma IHC. Immunohistochemistry staining of FFPE human prostate carcinoma tissue with Integrin Associated Protein Antibody clone rCD47/6365. HIER was performed by boiling tissue sections in pH 9 10mM Tris with 1mM EDTA for 20 min followed by cooling before testing. Strong brown staining is present throughout the malignant glandular epithelium, with prominent membranous and cytoplasmic labeling, while the PBS negative-control inset shows minimal background signal. This tumor-associated staining demonstrates tissue detection with the CD47 Recombinant Antibody.

Description

Integrin Associated Protein Antibody rCD47/6365 / CD47 Recombinant Antibody recognizes integrin-associated protein, a transmembrane glycoprotein encoded by the CD47 gene. CD47 was originally identified through its association with integrins and consequently became known as integrin-associated protein, or IAP. It is now recognized as a multifunctional cell-surface protein involved in immune recognition, cell adhesion, migration, and signal transduction. Important extracellular partners include signal regulatory protein alpha (SIRP alpha) and thrombospondin-1.

CD47 has an unusual membrane topology consisting of an extracellular immunoglobulin-like domain, five transmembrane segments, and a short alternatively spliced cytoplasmic tail. Through associations with integrins and other membrane proteins, CD47 participates in signaling complexes that influence adhesion, motility, survival, and cellular responses to the extracellular environment. The integrin-associated protein designation therefore remains relevant to understanding the discovery and broader biological functions of CD47.

One of the best-characterized functions of CD47 involves its interaction with SIRP alpha on myeloid cells. Engagement of SIRP alpha by CD47 can generate inhibitory signaling that restrains phagocytosis and contributes to mechanisms of self-recognition. This pathway has become an important area of cancer and immunology research because increased CD47 expression has been reported in numerous malignancies and may contribute to reduced macrophage-mediated clearance of tumor cells. An Integrin Associated Protein Antibody can support investigation of CD47 expression within this broader immune regulatory context.

CD47 also interacts with thrombospondin-1 and participates in biological processes extending beyond the CD47-SIRP alpha pathway. These interactions have linked CD47 with vascular regulation, cellular stress responses, adhesion, migration, and tissue homeostasis. The consequences of CD47 signaling can vary according to cellular context, ligand availability, and the composition of associated membrane complexes.

The names integrin-associated protein, IAP, CD47, OA3, and MER6 reflect different stages in the discovery and characterization of this membrane protein. NSJ Bioreagents offers clone rCD47/6365 as a recombinant monoclonal reagent for research into CD47 expression, immune recognition, cell adhesion, and receptor-associated signaling.

An Integrin Associated Protein Antibody such as clone rCD47/6365 provides a defined recombinant monoclonal reagent for investigating CD47 biology, while a CD47 Recombinant Antibody provides current cluster-of-differentiation terminology for studies of immune regulation, cell adhesion, and intercellular signaling.
