Recombinant His-Tag Chimeric Human Antibody Human HEK293 Cell WB. Western blot analysis was performed using lysates from human HEK293 cells transfected (+) or untransfected (-) with a DNA construct encoding a His-tagged Histone H3 protein. Recombinant His-Tag Chimeric Human Antibody clone RMH01 was used at 0.2 ug/mL, followed by an anti-human IgG secondary antibody for detection. A distinct immunoreactive band corresponding to the His-tagged Histone H3 protein is detected only in the transfected sample, while the untransfected control shows no detectable signal. These results demonstrate that Recombinant His-Tag Chimeric Human Antibody specifically detects His-tagged recombinant proteins and is fully compatible with standard anti-human IgG secondary antibody detection systems.
Recombinant His-Tag Chimeric Human Antibody Sandwich ELISA. A sandwich ELISA was performed using Recombinant His-Tag Chimeric Human Antibody clone RMH01 as the capture antibody at concentrations ranging from 0.125-2 ug/mL. Whole lysates from HEK293T cells expressing His-tagged Histone H3 K36M were added as the antigen, followed by an anti-Histone H3 antibody for detection. Absorbance was measured at 405 nm. Increasing concentrations of the capture antibody produced correspondingly stronger ELISA signals, demonstrating efficient capture of the His-tagged recombinant protein over a broad concentration range. These results show that Recombinant His-Tag Chimeric Human Antibody is well suited for sandwich ELISA applications using anti-human IgG-compatible detection systems.

Recombinant His-Tag Chimeric Human Antibody clone RMH01 was applied as the primary detection antibody at concentrations ranging from 0.008-0.5 ug/mL, followed by an HRP-conjugated anti-human IgG secondary antibody. Absorbance was measured at 405 nm. The antibody demonstrates strong, concentration-dependent binding to the His-tagged recombinant protein, with signal intensity decreasing predictably as both antigen and antibody concentrations are reduced. These results demonstrate that Recombinant His-Tag Chimeric Human Antibody provides sensitive and specific detection of His-tagged recombinant proteins in ELISA using standard anti-human IgG detection systems.

Recombinant His-Tag Chimeric Human Antibody is a recombinant rabbit-human chimeric monoclonal antibody developed for the highly specific detection of recombinant proteins containing a polyhistidine tag. Clone RMH01 was engineered by combining the antigen-binding variable regions of rabbit monoclonal clone RM146 with the constant region of human IgG1, producing a chimeric antibody that retains the excellent affinity and specificity of the original rabbit antibody while being recognized exclusively by anti-human IgG secondary antibodies. This unique design makes the antibody particularly valuable for assay development, secondary antibody validation, and experimental systems requiring a human IgG primary antibody. Recombinant His-Tag Chimeric Human Antibody recognizes recombinant proteins containing either a 6xHis tag or a 10xHis tag positioned at the amino terminus or the carboxy terminus of the target protein. The immunogen consisted of a mixture of peptides containing amino-terminal and carboxy-terminal 6xHis sequences, allowing clone RMH01 to recognize the His tag regardless of its orientation on the recombinant protein. Importantly, the antibody shows no detectable cross-reactivity with endogenous mammalian or bacterial proteins, providing highly selective detection of His-tagged recombinant proteins expressed in bacterial, yeast, insect, or mammalian expression systems.

Because the antibody carries a human IgG1 constant region, Recombinant His-Tag Chimeric Human Antibody is detected using anti-human IgG secondary antibodies rather than anti-rabbit reagents.
