PSGL-1 Antibody PSGL1/8192R / Leukocyte Trafficking Marker detects P-selectin glycoprotein ligand 1, a cell-surface glycoprotein encoded by the SELPLG gene and also known as CD162. PSGL-1 functions as a high-affinity counter-receptor for selectins and is expressed on myeloid cells, activated T lymphocytes and other leukocyte populations. Through interactions with P-, E- and L-selectin, PSGL-1 contributes to leukocyte adhesion and trafficking during inflammatory and immune responses.

PSGL-1 is a mucin-like transmembrane glycoprotein that undergoes extensive post-translational modification. High-affinity selectin binding requires modifications that include tyrosine sulfation and addition of the sialyl Lewis x carbohydrate structure to O-linked glycans. These modifications create functional selectin-binding sites and enable PSGL-1 to interact efficiently with selectins expressed on activated platelets and vascular endothelial cells.

Leukocyte trafficking is one of the principal functions associated with PSGL-1. During inflammation, interactions between CD162 on circulating leukocytes and selectins on activated vascular surfaces mediate initial tethering and rolling under blood flow. These transient adhesive interactions slow circulating leukocytes and facilitate subsequent firm adhesion and migration across the vascular endothelium. PSGL-1 is therefore widely studied in mechanisms controlling recruitment of immune cells to inflammatory sites.

PSGL-1 also contributes to immune biology beyond its mechanical role in cell adhesion. Selectin engagement can influence leukocyte activation and intracellular responses, while differences in PSGL-1 expression and glycosylation can regulate the ability of particular immune-cell populations to interact with vascular selectins. CD162 is consequently relevant to studies of innate and adaptive immunity, inflammatory recruitment and communication among leukocytes, platelets and endothelial cells.

Clone PSGL1/8192R is a recombinant rabbit monoclonal antibody generated against full-length recombinant human protein. It has been evaluated by immunohistochemistry in FFPE human tissues, with positive staining demonstrated in spleen and tonsil and human brain included as a negative-control tissue. Recombinant production provides a defined antibody sequence and supports consistent performance across manufacturing lots.
