PA2.26 Antigen Antibody PDPN/8950R in Human Lung WB. Western blot testing of human lung tissue lysate with PA2.26 Antigen Antibody / Podoplanin Glycoprotein Marker, clone PDPN/8950R, at 2-4 ug/ml. The predicted molecular weight of podoplanin is approximately 25 kDa, although the protein may be observed at higher molecular weights due to glycosylation. This PA2.26 Antigen Antibody demonstrates podoplanin detection in human lung tissue.
PA2.26 Antigen Antibody PDPN/8950R in Human Tonsil IHC. Immunohistochemistry staining of FFPE human tonsil tissue with PA2.26 Antigen Antibody / Podoplanin Glycoprotein Marker, clone PDPN/8950R, at 1-2 ug/ml for 30 min at RT. HIER was performed by boiling tissue sections in 10 mM Tris with 1 mM EDTA, pH 9, for 20 min followed by cooling before testing. PA2.26 Antigen Antibody / Podoplanin Glycoprotein Marker detects podoplanin (PDPN), a heavily glycosylated cell-surface protein also known as PA2.26 antigen, Gp36 and T1 alpha. Podoplanin is a mucin-type transmembrane glycoprotein expressed by lymphatic endothelial cells and several other specialized cell populations. Extensive post-translational glycosylation contributes to an observed molecular weight of approximately 38-43 kDa despite the smaller predicted molecular weight of the polypeptide.

Podoplanin is a type I transmembrane protein with a heavily O-glycosylated extracellular region and a short cytoplasmic tail. Although PDPN lacks intrinsic enzymatic activity, interactions involving its extracellular and intracellular domains can influence cell morphology, adhesion and migration. Podoplanin also binds the platelet receptor CLEC-2, an interaction important to platelet activation and the proper separation of developing blood and lymphatic vascular systems.

PA2.26 antigen is strongly associated with lymphatic endothelial biology. Podoplanin expression can be used to investigate lymphatic vessels, lymphatic endothelial differentiation and lymphangiogenesis. The protein is also expressed by stromal cells in peripheral lymphoid tissues and by thymic epithelial cells. Its distribution among lymphatic and specialized stromal cell populations makes PDPN relevant to studies of vascular development, tissue organization and cellular differentiation.

Podoplanin expression has also been investigated extensively in disease and cancer biology. PDPN is found in lymphangiomas, Kaposi sarcomas and subsets of angiosarcoma exhibiting lymphatic differentiation. It is additionally well established in studies of epithelioid mesothelioma and mesothelial differentiation. These broader associations connect the normal lymphatic and stromal biology of PA2.26 antigen with research into pathological tissue remodeling, tumor development and altered cellular phenotypes.

Clone PDPN/8950R is a recombinant rabbit monoclonal antibody generated against a recombinant fragment of human PDPN protein. It has been evaluated by Western blot and immunohistochemistry in human samples, including lung tissue lysate and FFPE tonsil tissue. Recombinant production provides a defined antibody sequence and supports consistent performance across manufacturing lots.
