CD18 Antibody recognizes integrin beta 2, a transmembrane adhesion receptor subunit encoded by the ITGB2 gene. CD18 is expressed predominantly by leukocytes and associates with several different alpha integrin subunits to form members of the beta 2 integrin family. These heterodimers include LFA-1 (CD11a/CD18), Mac-1 (CD11b/CD18), complement receptor 4 (CD11c/CD18), and alpha D beta 2 (CD11d/CD18). Because CD18 is the shared beta chain of these receptors, it has broad importance in leukocyte adhesion and immune cell function.
Integrin beta 2 is a type I transmembrane glycoprotein containing a large extracellular region, a single transmembrane segment, and a short cytoplasmic tail. Association with an alpha integrin chain creates a functional heterodimer capable of transmitting signals bidirectionally across the plasma membrane. Regulation of integrin conformation controls ligand affinity, while ligand engagement can initiate intracellular signaling and cytoskeletal changes that influence cell adhesion, spreading, and migration.
CD18-containing integrins interact with multiple ligands involved in leukocyte trafficking and immune responses. LFA-1 binds intercellular adhesion molecules such as ICAM1, while other beta 2 integrins recognize complement components and additional extracellular or cell-surface ligands. Through these interactions, ITGB2 contributes to leukocyte adhesion to vascular endothelium, transendothelial migration, immune synapse formation, phagocytosis, and inflammatory responses.
The physiological importance of ITGB2 is illustrated by leukocyte adhesion deficiency type I, which results from pathogenic variants affecting CD18 expression or function. Impaired beta 2 integrin activity compromises firm leukocyte adhesion and migration from the circulation into tissues, resulting in defective immune responses. ITGB2 and its associated integrins are therefore widely studied in leukocyte trafficking, innate and adaptive immunity, inflammation, cell adhesion, and immune disorders.
Although CD18 has historically been associated with the term LFA-1, CD18 itself is not unique to LFA-1. LFA-1 specifically consists of the CD11a alpha chain paired with CD18, whereas ITGB2 also forms heterodimers with CD11b, CD11c, and CD11d. Distinguishing the shared CD18 subunit from individual beta 2 integrin heterodimers is important when interpreting ITGB2 expression and function.
A CD18 Antibody can be used to investigate expression of the shared beta 2 integrin subunit, while an ITGB2 Polyclonal Antibody provides a polyclonal reagent for studies of leukocyte adhesion and beta 2 integrin biology.
For additional antibodies targeting cluster of differentiation proteins, see our CD Antibodies page.
The stated application concentrations are suggested starting amounts. Titration of the CD18 Antibody / ITGB2 Polyclonal Antibody may be required due to differences in protocols and secondary/substrate sensitivity.
Human partial recombinant protein (AA 404-769) was used as the immunogen for this CD18 Antibody.
After reconstitution, the CD18 Antibody can be stored for up to one month at 4oC. For long-term, aliquot and store at -20oC. Avoid repeated freezing and thawing.
