Ki-67 antigen (MKI67) is a nuclear proliferation-associated protein encoded by the MKI67 gene and widely used as a marker of actively cycling cells. Ki-67 Antibody for WB enables detection of the Ki-67 protein by western blot, allowing researchers to evaluate proliferative activity and cell cycle progression in cultured cells and tissue-derived lysates. Western blot detection of Ki-67 is frequently used to compare proliferation levels across experimental conditions, cancer cell lines, or treatment groups that alter cell cycle activity.

Ki-67 antibody, also referred to as Ki67 antibody or MKI67 antibody in the literature, recognizes a large nuclear protein expressed during the G1, S, G2, and mitotic phases of the cell cycle but largely absent in quiescent G0 cells. Because Ki-67 expression correlates closely with cellular proliferation, Ki-67 western blot analysis provides a reliable biochemical method for detecting actively proliferating cell populations. Researchers frequently monitor Ki-67 protein levels by western blot when studying cell cycle regulation, tumor growth, and responses to anti-proliferative therapies.

Ki-67 protein is unusually large and exhibits complex migration behavior during SDS-PAGE analysis. The predicted molecular weight of Ki-67 is approximately 320-360 kDa depending on isoform, and western blot analysis commonly reveals broad or multiple high molecular weight bands in this region. These band patterns arise from extensive phosphorylation and other post-translational modifications that occur as Ki-67 participates in chromatin organization during the cell cycle. As a result, Ki-67 western blot signals often appear as diffuse or doublet bands at very high molecular weight rather than a single sharp band.

Western blot detection of Ki-67 is particularly useful in studies examining proliferative signaling pathways and tumor biology. Increased Ki-67 protein levels detected by western blot often correspond to rapidly dividing cells, including many cancer cell lines. In contrast, differentiated or growth-arrested cells typically show reduced Ki-67 expression. Because of this strong association with cellular proliferation, Ki-67 protein detection by western blot is widely used alongside imaging-based approaches such as immunohistochemistry or immunofluorescence to evaluate proliferative status.
