Integrin Beta 2 Antibody 534CT6.2.1 recognizes integrin beta 2, a transmembrane adhesion receptor subunit encoded by the ITGB2 gene and commonly designated CD18. Integrin beta 2 is predominantly expressed by leukocytes and combines with several alpha integrin chains to form distinct heterodimeric receptors. These include LFA-1 (CD11a/CD18), Mac-1 (CD11b/CD18), complement receptor 4 (CD11c/CD18), and alpha D beta 2 (CD11d/CD18).
Integrin beta 2 is a type I transmembrane glycoprotein composed of a large extracellular region, a single transmembrane segment, and a short cytoplasmic tail. Association with different alpha chains generates receptors with distinct ligand-binding properties. Changes in integrin conformation regulate extracellular ligand affinity, while receptor engagement can initiate intracellular signaling and cytoskeletal rearrangements that influence leukocyte adhesion and migration.
Beta 2 integrins participate in interactions with intercellular adhesion molecules, complement components, and other extracellular or cell-surface ligands. Through these interactions, ITGB2-containing receptors contribute to firm leukocyte adhesion, transendothelial migration, immune synapse formation, phagocytosis, inflammatory responses, and other functions of innate and adaptive immune cells.
CD18 is the common beta subunit of the beta 2 integrin family rather than a component unique to LFA-1. LFA-1 specifically contains CD11a paired with CD18, whereas other alpha chains pair with the same ITGB2 protein to produce additional leukocyte integrins. This shared-subunit biology is important when interpreting CD18 expression because detection of integrin beta 2 does not by itself identify which alpha/beta heterodimer is present.
Pathogenic variants affecting ITGB2 can cause leukocyte adhesion deficiency type I, a disorder characterized by impaired beta 2 integrin expression or function. Disruption of CD18-dependent adhesion interferes with normal leukocyte recruitment from the circulation into tissues, illustrating the importance of beta 2 integrins in immune surveillance and host defense.
