IL7RA Antibody 22I71 Staining in Human Tonsil Lymphoid Tissue IHC. Immunohistochemistry staining of FFPE human tonsil tissue using IL7RA Antibody (clone 22I71) shows prominent brown staining in numerous lymphoid cells, with heterogeneous signal intensity across the tissue. Staining is predominantly membranous and cytoplasmic, while intervening areas contain comparatively less signal. HIER was performed by boiling tissue sections in pH 8 EDTA for 20 min followed by cooling before testing. This Interleukin 7 Receptor Alpha Antibody demonstrates detection of IL7RA/CD127 in human tonsil tissue.
IL7RA Antibody 22I71 Detects CD127 in Human, Rat and Mouse Samples WB. Western blot analysis using IL7RA Antibody (clone 22I71) shows strong, discrete bands migrating at approximately 65-70 kDa. Lane 1: human HeLa; lane 2: human Jurkat; lane 3: human HepG2; lane 4: human MCF7; lane 5: rat spleen; lane 6: rat PC-12; lane 7: mouse spleen; lane 8: mouse RAW264.7 cell lysate. Although the predicted molecular weight is approximately 52 kDa, IL7RA is glycosylated and can migrate at a higher apparent molecular weight. This Interleukin 7 Receptor Alpha Antibody produces a consistent, predominantly single-band pattern across all three species.

Description

IL7RA Antibody 22I71 recognizes interleukin 7 receptor alpha, a type I transmembrane glycoprotein encoded by the IL7R gene and also known as CD127. IL7RA serves as a receptor component for two cytokine systems with important roles in immune development and regulation. Association of IL7RA with the common gamma chain, IL2RG/CD132, forms the functional receptor for interleukin 7 (IL-7), while association with CRLF2 forms the receptor for thymic stromal lymphopoietin (TSLP).

IL-7 binding to its receptor activates JAK1 and JAK3 and promotes downstream signaling through STAT proteins, particularly STAT5. Additional pathways including PI3K-AKT contribute to cellular survival, proliferation, differentiation, and metabolism. Through these signaling mechanisms, IL7RA plays an important role in lymphocyte development and in the maintenance of mature lymphocyte populations.

Expression of IL7RA varies according to immune-cell lineage, differentiation, and activation state. CD127 is particularly important in T-cell biology and contributes to the development and maintenance of naive and memory T-cell populations. The receptor is also widely used as an immune-cell marker, including in studies that distinguish regulatory T cells from conventional T-cell populations. Altered IL7RA expression or signaling is studied in immune deficiency, autoimmunity, inflammation, leukemia, and other disorders involving lymphocyte development or homeostasis.

Clone 22I71 is a recombinant rabbit monoclonal antibody generated using a peptide sequence specific to interleukin 7 receptor alpha. As an Interleukin 7 Receptor Alpha Antibody, clone 22I71 provides a defined recombinant reagent for investigating IL7RA and its relationship to cytokine signaling and lymphocyte biology.
