IBA1 Antibody HuProt Validated clone AIF1/1909 recognizes Allograft inflammatory factor 1, a 17 kDa cytoplasmic EF-hand calcium-binding protein encoded by the AIF1 gene on chromosome 6p21.3 within the major histocompatibility complex region. Allograft inflammatory factor 1 is most widely known as IBA1, a well-established marker of microglia and macrophages in both central nervous system and peripheral immune tissues. IBA1 Antibody HuProt Validated clone AIF1/1909 is designed to detect endogenous IBA1 protein expression in research settings where high specificity is essential.
Allograft inflammatory factor 1, commonly referred to as IBA1 antibody target protein in the literature, is a member of the EF-hand superfamily and contains two calcium-binding EF-hand motifs that regulate actin cytoskeletal dynamics. The protein is primarily localized to the cytoplasm and cellular processes of microglia and tissue macrophages, where it participates in membrane ruffling, phagocytosis, migration, and inflammatory signaling. IBA1 expression is strongly upregulated during microglial activation and in response to neural injury, infection, ischemia, and neurodegenerative disease.
IBA1 is highly expressed in resident microglia of the brain and spinal cord and in macrophage populations within spleen, lymph node, tonsil, and inflamed peripheral tissues. During development, microglial precursors begin expressing IBA1 as they colonize the embryonic brain, and expression persists throughout adulthood. In pathological contexts, increased IBA1 expression correlates with reactive microgliosis in disorders such as Alzheimer's disease, multiple sclerosis, traumatic brain injury, and brain tumors.
HuProt microarray validation of clone AIF1/1909 demonstrates strong target specificity against full-length human AIF1 among more than 19,000 proteins tested. A portion of amino acids 1-146 from the human protein was used as the immunogen for the IBA1 antibody.
