Immunohistochemistry of IBA1 antibody in human lymph node. Formalin-fixed, paraffin-embedded human lymph node tissue shows strong cytoplasmic HRP-DAB brown staining in macrophages and dendritic-like cells within the paracortical and sinusoidal regions. Staining was performed using clone AIF1/1909, a HuProt validated antibody, following heat-induced epitope retrieval by boiling sections in pH 6 citrate buffer for 10-20 min and cooling at room temperature for 20 min. The staining pattern is consistent with expected IBA1 expression in tissue-resident macrophages.
Immunohistochemistry of IBA1 antibody in human kidney tissue. Formalin-fixed, paraffin-embedded human kidney shows cytoplasmic HRP-DAB brown staining in interstitial macrophages and mononuclear cells surrounding renal tubules and glomeruli, while tubular epithelial cells remain largely negative. Staining was performed using clone AIF1/1909, a HuProt validated antibody, following heat-induced epitope retrieval by boiling sections in pH 6 citrate buffer for 10-20 min and cooling at room temperature for 20 min. The staining distribution is consistent with expected IBA1 expression in tissue-resident macrophages.
Immunohistochemistry of IBA1 antibody in human tonsil. Formalin-fixed, paraffin-embedded human tonsil tissue shows cytoplasmic HRP-DAB brown staining in scattered macrophages and dendritic-like cells within the interfollicular regions and germinal centers, with lymphocytes largely negative.
Human protein microarray specificity validation of IBA1 antibody. Analysis of HuProt(TM) microarray containing more than 19,000 full-length human proteins was performed using IBA1 antibody HuProt validated clone AIF1/1909. The antibody demonstrates strongest binding to AIF1 with a Z-score of 94.75 and an S-score of 81.96, indicating clear separation from all other proteins on the array. These results support the high specificity of clone AIF1/1909 for its intended target.
Z- and S-score explanation: The Z-score represents the strength of signal produced when the antibody, in combination with a fluorescently tagged anti-IgG secondary antibody, binds to a specific protein on the HuProt(TM) array. Z-scores are expressed in standard deviations above the mean signal of all proteins on the array. Proteins are ranked in descending order based on Z-score. The S-score represents the difference in Z-scores between sequentially ranked proteins and reflects the relative specificity of the antibody for its intended target compared to potential off-target interactions.

Description

IBA1 Antibody HuProt Validated clone AIF1/1909 recognizes Allograft inflammatory factor 1, a 17 kDa cytoplasmic EF-hand calcium-binding protein encoded by the AIF1 gene on chromosome 6p21.3 within the major histocompatibility complex region. Allograft inflammatory factor 1 is most widely known as IBA1, a well-established marker of microglia and macrophages in both central nervous system and peripheral immune tissues. IBA1 Antibody HuProt Validated clone AIF1/1909 is designed to detect endogenous IBA1 protein expression in research settings where high specificity is essential.

Allograft inflammatory factor 1, commonly referred to as IBA1 antibody target protein in the literature, is a member of the EF-hand superfamily and contains two calcium-binding EF-hand motifs that regulate actin cytoskeletal dynamics. The protein is primarily localized to the cytoplasm and cellular processes of microglia and tissue macrophages, where it participates in membrane ruffling, phagocytosis, migration, and inflammatory signaling. IBA1 expression is strongly upregulated during microglial activation and in response to neural injury, infection, ischemia, and neurodegenerative disease.

IBA1 is highly expressed in resident microglia of the brain and spinal cord and in macrophage populations within spleen, lymph node, tonsil, and inflamed peripheral tissues. During development, microglial precursors begin expressing IBA1 as they colonize the embryonic brain, and expression persists throughout adulthood. In pathological contexts, increased IBA1 expression correlates with reactive microgliosis in disorders such as Alzheimer's disease, multiple sclerosis, traumatic brain injury, and brain tumors.

HuProt microarray validation of clone AIF1/1909 demonstrates strong target specificity against full-length human AIF1 among more than 19,000 proteins tested. Z-score and S-score metrics indicate clear separation of the intended target from off-target proteins, supporting the specificity profile of this IBA1 Antibody HuProt Validated clone. Such high-density protein array validation provides an orthogonal specificity assessment beyond tissue staining alone.

Given its established role in innate immune activation and cytoskeletal remodeling, IBA1 serves as a critical research marker for microglial density, morphology, and activation state. A well-characterized IBA1 Antibody HuProt Validated clone AIF1/1909 enables investigation of neuroinflammation, macrophage infiltration, and immune responses in both normal and disease-associated tissues. This antibody targets IBA1 in research applications requiring strong target selectivity.
