HSP60 Antibody AE-1 / Heat Shock 60 kDa Protein Antibody in HeLa Cervical Cancer Cells IF. Immunofluorescence staining of PFA-fixed human HeLa cervical cancer cells with HSP60 Antibody clone AE-1 shows strong granular cytoplasmic staining (green) surrounding the nuclei, consistent with the predominantly mitochondrial localization of HSP60. RedDot nuclear staining (red) identifies the nuclei and highlights the extranuclear distribution of HSP60.
HSP60 Antibody AE-1 in Human Renal Cell Carcinoma IHC. Immunohistochemistry staining of FFPE human renal cell carcinoma tissue with HSP60 Antibody clone AE-1 shows prominent granular cytoplasmic staining within tumor cells, consistent with mitochondrial HSP60 localization. Heat-induced epitope retrieval was performed by boiling tissue sections in pH 9 10 mM Tris with 1 mM EDTA for 20 min followed by cooling before testing.

Description

HSP60 Antibody AE-1 / Heat Shock 60 kDa Protein Antibody recognizes HSP60, a highly conserved molecular chaperone encoded by the HSPD1 gene. HSP60 is predominantly localized to the mitochondrial matrix, where it participates in the folding and assembly of proteins required for mitochondrial function. Together with its co-chaperonin HSP10, HSP60 forms an ATP-dependent chaperone system that helps maintain mitochondrial protein homeostasis.

HSP60 is initially synthesized as a precursor containing an N-terminal mitochondrial targeting sequence. After import into mitochondria, this sequence is removed to produce the mature protein. HSP60 subunits assemble into oligomeric complexes that provide a protected environment for unfolded and partially folded proteins, allowing them to acquire functional conformations while limiting inappropriate protein interactions.

Expression and function of HSP60 can be influenced by environmental and physiological stress. In addition to its essential role in mitochondrial proteostasis, HSPD1 has been studied in oxidative stress, mitochondrial dysfunction, inflammation, immune responses, neurodegeneration, and cancer. Altered abundance or localization of HSP60 may accompany disturbances in cellular homeostasis, making the protein relevant to research across mitochondrial biology and disease-associated processes.

HSP60 is also of interest in immunology because mammalian HSP60 is evolutionarily related to bacterial chaperonins. Immune responses directed against bacterial HSP60 proteins may recognize conserved features shared with mammalian homologs, and HSP60-associated immune reactivity has been investigated in inflammatory and autoimmune disease. These relationships connect Heat Shock 60 kDa Protein research with cellular stress, mitochondrial biology, microbiology, and immunology.
