HBA2 Antibody / Hemoglobin Subunit Alpha 2 Antibody recognizes alpha-globin encoded by the HBA2 gene. HBA2 is one of two closely related functional alpha-globin genes located within the alpha-globin gene cluster on human chromosome 16. HBA2 and HBA1 encode identical alpha-globin protein sequences, although the two genes differ in their relative contribution to total alpha-globin production. Alpha-globin is a fundamental component of hemoglobin and is highly expressed during erythroid differentiation and red blood cell development.
Alpha-globin associates with beta-like globin chains to form functional hemoglobin tetramers. In adults, alpha chains pair predominantly with beta-globin to form hemoglobin A (HbA; alpha2beta2), the major hemoglobin responsible for oxygen transport in circulating erythrocytes. Alpha-globin also associates with delta-globin to form HbA2, while during fetal development it pairs with gamma-globin to form fetal hemoglobin. These developmentally regulated partnerships allow alpha-globin to remain an essential component of hemoglobin throughout much of human development and adult life.
Proper regulation of HBA2 and the neighboring HBA1 gene is required to maintain balanced globin-chain synthesis. Deletions and other pathogenic alterations affecting the alpha-globin locus can reduce alpha-chain production and cause alpha-thalassemia. Disease severity depends on the number and functional status of affected alpha-globin genes, ranging from relatively mild hematologic changes to severe forms of alpha-thalassemia. Alpha-globin biology is therefore studied extensively in erythropoiesis, hemoglobinopathies, developmental hematology and red blood cell research.
