Granzyme B Antibody recognizes granzyme B (GZMB), a cytotoxic serine protease that serves as one of the principal effector molecules released by activated CD8-positive cytotoxic T lymphocytes and natural killer (NK) cells during cell-mediated immune responses. Following immune synapse formation, granzyme B enters target cells through perforin-generated pores and initiates apoptosis by activating caspases and cleaving multiple intracellular substrates, including BID and PARP. This highly regulated mechanism is essential for eliminating virus-infected, transformed, and malignant cells, making Granzyme B Antibody an important research tool for investigating immune surveillance, host defense, and cytotoxic lymphocyte function.

Granzyme B expression is largely restricted to activated cytotoxic immune cells and is therefore widely used as a functional biomarker of immune activation rather than simply a marker of lymphocyte lineage. Increased GZMB expression has been associated with effective antitumor immunity, antiviral responses, transplant rejection, autoimmune disease, and chronic inflammatory disorders. Consequently, Granzyme B Antibody is frequently used to identify activated cytotoxic T cells and NK cells within tissues and to evaluate immune responses in oncology, infectious disease, and immunology research.

This recombinant rabbit monoclonal Granzyme B Antibody (clone RM441) was generated against a peptide corresponding to the C-terminal region of granzyme B. Because the C-terminus is retained following proteolytic maturation of the protein, antibodies directed against this region are expected to recognize both precursor and mature forms of GZMB while maintaining high target specificity.
