EWS Antibody HeLa Nuclear WB. Western blot analysis of nuclear HeLa cell lysate using EWS Antibody at 0.3 ug/ml. A single prominent band is detected at approximately 90 kDa. Although the predicted molecular weight of EWS is approximately 69 kDa, the protein routinely migrates at a higher apparent molecular weight because of its intrinsically disordered domains and unusual amino acid composition, which alter electrophoretic mobility during SDS-PAGE. These results demonstrate that this EWS Antibody, an Ewing Sarcoma Protein Antibody, specifically detects endogenous nuclear EWS and is suitable for Western blot analysis.
EWS Antibody Human Kidney IHC. Immunohistochemical analysis of FFPE human kidney tissue following heat-induced epitope retrieval in pH6 citrate buffer. Tissue sections were incubated with EWS Antibody at 7 ug/ml, followed by an HRP-conjugated secondary antibody and chromogenic detection. Predominantly nuclear staining is observed throughout renal tubular epithelial cells, with minimal staining in glomeruli, consistent with the known nuclear localization of EWS as an RNA-binding protein involved in transcriptional regulation and RNA processing. This staining pattern demonstrates that this EWS Antibody, an Ewing Sarcoma Protein Antibody, is suitable for detecting endogenous EWS expression in human FFPE kidney tissue by immunohistochemistry.
EWS Antibody Human Kidney Negative Control IHC. Negative control immunohistochemical analysis of FFPE human kidney tissue following heat-induced epitope retrieval in pH6 citrate buffer. Tissue sections were processed using the same HRP staining protocol as the experimental sample but without the primary antibody. No specific DAB staining is observed, demonstrating minimal background signal and supporting the specificity of the EWS Antibody staining observed in the corresponding human kidney tissue section.

Description

EWS Antibody / Ewing Sarcoma Protein Antibody recognizes EWS, also known as Ewing Sarcoma Protein or EWSR1, a multifunctional RNA-binding protein that belongs to the FET family alongside FUS and TAF15. EWS is widely expressed in mammalian tissues and serves as an important regulator of transcription, RNA processing, messenger RNA transport, and genome stability. The protein contains an amino-terminal transcriptional activation domain and a carboxyl-terminal RNA-binding domain, enabling interactions with both transcriptional machinery and RNA molecules. Because of its central role in coordinating gene expression, an EWS Antibody is widely used to investigate nuclear RNA metabolism and transcriptional regulation in both normal physiology and disease.

EWS functions as a molecular scaffold that links transcription with multiple post-transcriptional events, including alternative RNA splicing and messenger RNA maturation. It interacts with RNA polymerase II, numerous RNA-binding proteins, and components of the spliceosome to coordinate efficient gene expression. In addition, EWS participates in cellular responses to DNA damage and contributes to the formation of dynamic nuclear condensates through liquid-liquid phase separation. These diverse activities have established EWS as an important regulator of cellular homeostasis and have made an EWS Antibody a valuable tool for studies of RNA biology, nuclear organization, and gene regulation.

The greatest clinical significance of EWS stems from recurrent chromosomal translocations involving the EWSR1 gene. The resulting fusion proteins, particularly EWSR1-FLI1, function as potent oncogenic transcriptional regulators and represent the defining molecular hallmark of Ewing sarcoma. Additional EWSR1 fusion proteins occur in several other soft tissue tumors and sarcomas, including clear cell sarcoma, desmoplastic small round cell tumor, extraskeletal myxoid chondrosarcoma, and myxoid liposarcoma. Beyond cancer, alterations in EWS function have been linked to defects in RNA metabolism, DNA repair, and cellular stress responses, expanding interest in the protein across multiple areas of biomedical research. Consequently, an EWS Antibody is frequently used in molecular pathology, cancer biology, and RNA regulation studies.

EWS is commonly analyzed by Western blot, immunohistochemistry, immunofluorescence, immunoprecipitation, and flow cytometry to evaluate protein expression, localization, and molecular interactions. These complementary techniques allow investigators to examine EWS function in cultured cells, normal tissues, and tumor specimens while supporting studies of transcriptional regulation and oncogenic gene fusions. NSJ Bioreagents provides carefully validated antibodies to facilitate these applications across diverse experimental models. An EWS Antibody / Ewing Sarcoma Protein Antibody is a valuable research reagent for investigating RNA-binding proteins, transcriptional control, EWSR1-associated malignancies, and the molecular mechanisms governing gene expression.
