Cytokeratin 6 Antibody KRT6A/2368 recognizes keratin 6A (KRT6A), a type II intermediate filament protein involved in epithelial differentiation, tissue integrity, and responses to injury. Also known as cytokeratin 6A or CK6, this approximately 60 kDa protein is encoded by the KRT6A gene on chromosome 12. KRT6A is expressed in selected stratified epithelia, including hair follicles, nail beds, and specialized regions of the oral mucosa. Its expression can increase substantially in activated keratinocytes during wound healing, inflammation, and epithelial hyperproliferation, making it a useful marker of changes in epithelial differentiation.
KRT6A forms heterodimers with type I keratins, particularly keratins 16 and 17, to assemble intermediate filament networks that support cellular architecture. Unlike the KRT5/KRT14 pair associated with basal keratinocytes or the KRT1/KRT10 pair characteristic of differentiated epidermis, KRT6A is frequently associated with activated epithelial states. Its expression is induced in keratinocytes responding to mechanical injury, inflammatory signaling, and tissue remodeling. These changes help distinguish normal epithelial differentiation from the adaptive responses that occur during repair and disease.
Cytokeratin 6 is widely investigated in squamous cell carcinoma and other epithelial malignancies. KRT6A expression is frequently associated with squamous differentiation and can be evaluated alongside markers such as KRT5, KRT14, and p63. Increased KRT6A expression has also been reported in hyperproliferative skin disorders, including psoriasis, where altered keratin expression accompanies abnormal epidermal differentiation. Pathogenic variants in KRT6A are associated with pachyonychia congenita, an inherited disorder characterized by nail abnormalities, painful palmoplantar keratoderma, and other epithelial manifestations.
NSJ Bioreagents offers mouse monoclonal clone KRT6A/2368, developed against a recombinant human KRT6A fragment. The antibody is supported by immunohistochemistry, flow cytometry, and protein microarray analysis. HuProt screening identifies KRT6A as the strongest binding target, although additional signals indicate potential secondary interactions under the assay conditions. This distinction is important when interpreting antibody specificity across different experimental applications. A Cytokeratin 6 Antibody provides a useful reagent for investigating keratinocyte activation, epithelial differentiation, wound healing, and squamous epithelial tumor biology.
