IAP Antibody CD47/3019 / CD47 Monoclonal Antibody recognizes integrin-associated protein (IAP), a widely expressed transmembrane glycoprotein encoded by the CD47 gene. CD47 was originally identified through its association with integrins and was consequently named IAP. Subsequent research established CD47 as a multifunctional cell-surface protein involved in immune recognition, cell adhesion, migration, and signal transduction. Its extracellular binding partners include signal regulatory protein alpha (SIRP alpha) and thrombospondin-1, connecting CD47 to several distinct biological pathways.
CD47 has an unusual membrane organization consisting of an extracellular immunoglobulin-like domain, five transmembrane segments, and a short alternatively spliced cytoplasmic tail. Its association with integrins and other membrane proteins allows CD47 to participate in multiprotein signaling complexes that regulate cellular adhesion, motility, survival, and responses to the extracellular environment. The historical IAP designation therefore remains biologically relevant because integrin-associated functions represent an important component of CD47 biology.
A major area of CD47 research concerns its interaction with SIRP alpha. Engagement of SIRP alpha on myeloid cells by CD47 can initiate inhibitory signaling that restrains phagocytosis, contributing to recognition of cells as self. This mechanism has attracted substantial interest in cancer biology because increased CD47 expression has been reported in numerous malignancies and may reduce macrophage-mediated elimination of tumor cells. An IAP Antibody can therefore support studies examining CD47 abundance and the cellular mechanisms associated with immune recognition.
CD47 also binds thrombospondin-1 and participates in pathways extending beyond the CD47-SIRP alpha axis. 1. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
