CD41a Antibody ITGA2B/1036 recognizes CD41a, a platelet-associated antigen encoded by the ITGA2B gene and also known as integrin alpha 2b and platelet glycoprotein IIb. Integrin alpha 2b associates with integrin beta 3, or CD61, to form the alpha IIb beta 3 heterodimeric receptor. This complex is historically known as glycoprotein IIb/IIIa and is highly expressed on platelets and megakaryocytes. Clone ITGA2B/1036 recognizes a calcium-dependent CD41/CD61 complex, providing a distinct reagent for investigating this functionally important platelet surface receptor.
The CD41/CD61 complex has a central role in platelet adhesion and aggregation. In resting platelets, alpha IIb beta 3 is maintained primarily in a low-affinity state. Platelet activation induces conformational changes that increase receptor affinity for fibrinogen and other adhesive ligands. Fibrinogen can subsequently bridge receptors on neighboring platelets, promoting aggregation and formation of the primary hemostatic plug. Ligand engagement also initiates outside-in signaling that contributes to cytoskeletal reorganization, platelet spreading and stabilization of the developing clot.
CD41a is strongly associated with the megakaryocytic lineage and is widely used as a platelet marker in hematopoietic research. Expression of the CD41/CD61 complex appears during megakaryocyte maturation and is maintained on circulating platelets. CD41 is therefore useful for studying megakaryocytic differentiation, platelet development and hematopoietic cell populations. Pathogenic alterations affecting ITGA2B can impair alpha IIb beta 3 expression or activity and are associated with Glanzmann thrombasthenia, an inherited bleeding disorder characterized by defective platelet aggregation.
Clone ITGA2B/1036 was generated against recombinant full-length human integrin alpha IIb and recognizes the calcium-dependent CD41/CD61 complex. This characteristic differentiates the antibody from reagents directed simply against an isolated linear region of ITGA2B and makes the Platelet Marker Antibody particularly relevant to studies of the native platelet-associated receptor complex.
