BSEP Antibody Human Liver IHC. Immunohistochemical staining of FFPE human liver tissue using BSEP Antibody / Bile Salt Export Pump Antibody at 2 ug/ml. HIER was performed in pH 9 Tris-EDTA buffer. An HRP-conjugated secondary antibody with DAB substrate was used for detection. BSEP Antibody demonstrates strong canalicular membranous staining of hepatocytes in a characteristic chicken-wire pattern, consistent with the normal localization of the bile salt export pump in liver tissue.

BSEP Antibody recognizes the bile salt export pump (BSEP), also known as ATP-binding cassette subfamily B member 11 (ABCB11), a liver-specific ATP-dependent transporter responsible for the secretion of bile acids from hepatocytes into the bile canaliculi. Localized almost exclusively to the canalicular membrane of hepatocytes, BSEP plays an essential role in maintaining bile flow, cholesterol metabolism, and enterohepatic circulation. By exporting bile salts against a concentration gradient, BSEP helps preserve hepatocellular homeostasis while facilitating the digestion and absorption of dietary lipids and fat-soluble vitamins. Because of its highly restricted hepatic expression, BSEP Antibody is widely used to investigate liver physiology, hepatocyte polarity, and bile acid transport.

Loss of BSEP function disrupts normal bile acid secretion, leading to intracellular accumulation of toxic bile salts and progressive liver injury. Mutations in the ABCB11 gene are responsible for several inherited cholestatic disorders, including progressive familial intrahepatic cholestasis type 2 (PFIC2), benign recurrent intrahepatic cholestasis type 2 (BRIC2), and other forms of hereditary cholestasis. Reduced BSEP expression or impaired transporter activity has also been associated with drug-induced liver injury, pregnancy-associated cholestasis, chronic liver disease, and hepatocellular carcinoma. Consequently, BSEP Antibody is an important tool for studies of hepatic transporter biology, cholestatic disease mechanisms, and liver pathology.

Beyond inherited disease, BSEP has become an important target during pharmaceutical development because inhibition of the transporter is a recognized mechanism underlying drug-induced cholestasis. Many compounds are routinely screened for their effects on BSEP activity during preclinical safety assessment to reduce the risk of hepatotoxicity. Immunohistochemical analysis of BSEP expression is also valuable for evaluating hepatocyte differentiation, canalicular integrity, and biliary function in both experimental models and clinical tissue specimens. The characteristic canalicular chicken-wire staining pattern observed with BSEP Antibody makes it a useful marker of normal hepatocyte polarity and canalicular membrane architecture.
