ATG7 Antibody Human Pancreatic Cancer IHC. Immunohistochemistry was performed on paraffin-embedded human pancreatic cancer tissue using ATG7 Antibody at 2 ug/ml. Heat-induced epitope retrieval was carried out in pH 8 EDTA buffer, and the tissue was incubated with the primary antibody overnight at 4°C. Bound antibody was detected using an HRP-conjugated goat anti-rabbit IgG secondary antibody and DAB chromogen. Distinct cytoplasmic staining is observed throughout the pancreatic carcinoma cells, while the surrounding stromal tissue exhibits comparatively low background staining. These results demonstrate that this Autophagy Related E1 Enzyme Antibody is suitable for immunohistochemical detection of ATG7 in formalin-fixed, paraffin-embedded human pancreatic cancer tissue.
ATG7 Antibody A549 Cell IF. Immunofluorescence was performed on human A549 cells using ATG7 Antibody at 5 ug/ml. Cells underwent enzyme antigen retrieval for 15 minutes prior to incubation with the primary antibody overnight at 4°C. Bound antibody was detected using a Fluoro488-conjugated goat anti-rabbit IgG secondary antibody (green), and nuclei were counterstained with DAPI (blue). Diffuse cytoplasmic staining is observed throughout the A549 cells with minimal nuclear signal, consistent with the expected intracellular localization of ATG7. Lane 1: human HepG2 whole cell lysate; lane 2: human PC-3 whole cell lysate; lane 3: rat spleen tissue lysate; lane 4: mouse spleen tissue lysate. Proteins were separated on a 10% SDS-PAGE gel, transferred to a nitrocellulose membrane, and the blot was incubated with the primary antibody overnight at 4°C. Bound antibody was detected using an HRP-conjugated goat anti-rabbit IgG secondary antibody and enhanced chemiluminescence. A specific immunoreactive band is detected at approximately 78 kDa in all tested samples, consistent with the expected molecular weight of ATG7. ATG7 Antibody HepG2 Cell FACS. Flow cytometric analysis was performed on fixed and permeabilized human HepG2 cells using ATG7 Antibody at 1 ug/10^6 cells. The red histogram represents the unstained blank control, the green histogram represents the rabbit IgG isotype control, and the blue histogram represents cells stained with the ATG7 antibody. A clear rightward shift of the ATG7-stained population relative to both control populations demonstrates specific intracellular antibody binding and confirms ATG7 expression in HepG2 cells.
