AR Antibody / Androgen Nuclear Receptor Antibody recognizes androgen receptor, a ligand-activated transcription factor encoded by the AR gene. Also designated NR3C4, androgen receptor belongs to the nuclear receptor superfamily and mediates cellular responses to androgenic hormones, particularly testosterone and dihydrotestosterone. AR contains an N-terminal transcriptional regulatory domain, a central DNA-binding domain, a hinge region and a C-terminal ligand-binding domain. These structural regions coordinate hormone recognition, receptor activation, DNA binding and regulation of androgen-responsive genes.

Binding of an androgen ligand induces conformational changes in AR that influence receptor interactions, nuclear localization and transcriptional activity. Activated androgen receptor binds specific androgen response elements within regulatory regions of target genes and recruits transcriptional coregulators. Through these mechanisms, AR controls gene expression programs involved in cellular growth, differentiation, metabolism and survival. Its ability to translate steroid hormone signals into changes in transcription makes AR an important component of hormone-dependent cellular regulation.

Androgen receptor has essential roles in male sexual differentiation, reproductive development and maintenance of normal prostate physiology. AR signaling is also extensively investigated in prostate cancer, where receptor activity can continue to promote tumor growth and survival during disease progression. Mechanisms including AR amplification, mutation, altered coregulator activity and generation of alternatively spliced receptor variants can modify androgen signaling and contribute to treatment resistance. Outside oncology, pathogenic changes in AR are associated with androgen insensitivity syndromes and spinal and bulbar muscular atrophy, illustrating the broad physiological importance of the receptor.
