ZNF81 Antibody PCRP-ZNF81-2C7 / KRAB Zinc Finger Antibody recognizes zinc finger protein 81 (ZNF81), a member of the Krueppel C2H2-type zinc finger protein family. ZNF81 is a transcriptional regulator containing 12 C2H2-type zinc finger motifs and a Krueppel-associated box (KRAB) domain. C2H2 zinc fingers provide sequence-specific nucleic acid-binding capabilities in many regulatory proteins, while the KRAB domain is a potent transcriptional repression module found in a large family of mammalian zinc finger proteins.

KRAB zinc finger proteins commonly regulate gene expression through cooperation with transcriptional corepressors. The KRAB domain can interact with KAP1, also known as TRIM28, which functions as a scaffold for chromatin-regulatory factors. KAP1-associated complexes can recruit proteins involved in histone modification and chromatin remodeling, including SETDB1, heterochromatin protein 1, and histone deacetylase-containing complexes. These interactions provide a mechanism through which KRAB zinc finger proteins can promote transcriptional silencing and influence local chromatin states.

ZNF81 is encoded on the X chromosome and has historically also been referred to as HFZ20 and MRX45. Genetic alterations involving ZNF81 have been investigated in relation to X-linked neurodevelopmental phenotypes, making the protein relevant to research examining transcriptional regulation and nervous system development. Although the specific genomic targets and regulatory functions of ZNF81 remain less extensively characterized than those of some other KRAB zinc finger proteins, its domain architecture places it within a major family of proteins involved in transcriptional and chromatin regulation.
