YAP1 Antibody recognizes Yes-associated protein 1 (YAP1), a transcriptional coactivator and major downstream effector of the Hippo signaling pathway. Originally identified through its interaction with the Src family kinase YES1, YAP1 lacks intrinsic DNA-binding activity and instead regulates gene expression through interactions with TEAD transcription factors and other partners. Hippo pathway activation results in phosphorylation and cytoplasmic sequestration of YAP1, whereas pathway inhibition permits its translocation to the nucleus, where it activates genes involved in cell proliferation, survival, differentiation, and tissue regeneration. Tight control of YAP1 activity is essential for maintaining tissue homeostasis and organ size.
YAP1 is broadly expressed in embryonic and adult tissues and functions as a mechanosensor that responds to cell density, extracellular matrix stiffness, and mechanical stress. Through crosstalk with Wnt, transforming growth factor beta, epidermal growth factor receptor, phosphoinositide 3 kinase AKT, and G protein coupled receptor signaling pathways, YAP1 integrates diverse extracellular cues to coordinate cellular growth and differentiation. YAP1 activity is particularly important in stem and progenitor cell populations, where it contributes to self renewal and lineage specification. Experimental models have demonstrated essential roles for YAP1 in heart development, liver growth, wound healing, and tissue regeneration.
Abnormal YAP1 activation has been implicated in numerous diseases, especially cancer. Elevated YAP1 expression or nuclear localization has been observed in hepatocellular carcinoma, pancreatic cancer, breast cancer, lung cancer, colorectal cancer, ovarian cancer, gastric cancer, and glioblastoma. Increased YAP1 signaling promotes epithelial mesenchymal transition, angiogenesis, metastasis, cancer stem cell maintenance, and resistance to therapy. Beyond oncology, YAP1 has also been linked to fibrosis, cardiovascular disease, and inflammatory disorders. Because of its central role in growth control and cellular plasticity, YAP1 has emerged as an important biomarker and therapeutic target, making YAP1 Antibody a valuable tool for studies of Hippo signaling, tissue regeneration, and disease pathogenesis.
