TNC Antibody TNC/4806 / Hexabrachion Antibody recognizes Tenascin C, a large extracellular matrix glycoprotein encoded by the TNC gene. Also historically known as hexabrachion and cytotactin, Tenascin C is a disulfide-linked hexameric protein that contributes to extracellular matrix organization and regulates interactions between cells and their surrounding environment. Its modular structure includes epidermal growth factor-like repeats, fibronectin type III domains, and a fibrinogen-like region that support interactions with matrix proteins, cell-surface receptors, and other extracellular molecules.
Tenascin C expression is highly regulated during development. It is particularly associated with regions undergoing active tissue remodeling and mesenchymal-epithelial interactions, where it can influence cell adhesion, migration, proliferation, and differentiation. Expression becomes more restricted in normal adult tissues but can increase substantially during wound healing, inflammation, fibrosis, and other processes involving extracellular matrix remodeling.
TNC is also widely studied in cancer biology because Tenascin C can become strongly expressed within tumor-associated stroma and neovasculature. Increased expression has been reported in multiple tumor types and has been investigated in connection with tumor cell invasion, angiogenesis, metastatic progression, and changes within the tumor microenvironment. Rather than serving solely as a structural matrix component, Tenascin C can modify cell-matrix interactions and signaling processes that influence cellular behavior.
Alternative splicing of TNC produces multiple protein isoforms, particularly through variation within its fibronectin type III repeat region. This structural diversity contributes to differences in molecular size and may influence the biological properties and distribution of individual Tenascin C forms.
