TM4SF4 Antibody by IHC in human colon cancer tissue. Immunohistochemistry of paraffin-embedded human colon cancer tissue using Transmembrane 4 L6 family member 4 Antibody. Brown DAB staining is distributed among tumor cells, with blue nuclear counterstaining. Heat-mediated antigen retrieval used EDTA buffer at pH 8.0, followed by 10% goat serum blocking. The section was incubated with 2 ug/ml primary antibody overnight at 4oC, then treated with peroxidase-conjugated goat anti-rabbit IgG for chromogenic visualization.

Western blot analysis using Transmembrane 4 L6 family member 4 Antibody on six reducing lysates: A549, Caco-2, SH-SY5Y and U2OS human cells in lanes 1-4, followed by rat testis and mouse testis in lanes 5-6. A dominant band migrates near 40 kDa across the lanes, above the calculated unglycosylated molecular weight of approximately 21 kDa. TM4SF4 is a glycoprotein with reported glycosylated forms in the 25-40 kDa range, making glycosylation a plausible explanation for the higher migration. This blot alone does not establish the modification state.

Description

TM4SF4 Antibody / Transmembrane 4 L6 family member 4 Antibody targets a protein with important roles in cellular biology. Transmembrane 4 L6 family member 4 (TM4SF4) is a four-pass membrane protein belonging to the L6 family of tetraspan-like proteins. Its membrane-spanning architecture positions it to participate in interactions at the cell surface, where proteins coordinate adhesion, signaling and communication with the extracellular environment. Although it resembles tetraspanins in overall topology, TM4SF4 belongs to a distinct family and should not automatically be assigned every function established for classical tetraspanins.

TM4SF4 has been investigated in epithelial biology, including settings involving cell growth, differentiation and tissue remodeling. Its distribution within membranes and interactions with neighboring proteins may affect the organization of signaling assemblies. Studies in selected cancer models have reported associations between TM4SF4 expression and changes in proliferation or invasive behavior, but the direction and strength of those relationships can depend on cellular context. Expression differences alone do not establish that TM4SF4 drives a particular disease process.

Membrane proteins with several transmembrane helices can influence cellular responses through local protein organization rather than through intrinsic enzymatic activity. This distinction is relevant to TM4SF4, for which a catalytic function has not been established. Understanding its biological role requires identifying its binding partners, the conditions that regulate its abundance and the downstream pathways affected when its expression changes. Such work can also help separate functions shared with related L6-family proteins from those unique to TM4SF4.
