Immunofluorescence analysis of TBX5 using anti-TBX5 antibody. TBX5 expression was examined in a paraffin-embedded section of human lung cancer tissue. Heat-mediated antigen retrieval was performed using EDTA buffer (pH 8.0), followed by blocking with goat serum. The tissue section was incubated with a rabbit anti-TBX5 antibody overnight at 4°C, then stained with a Cy3-conjugated goat anti-rabbit secondary antibody. Nuclei were counterstained with DAPI. Fluorescent signals were visualized by fluorescence microscopy, revealing prominent nuclear localization of TBX5 consistent with its role as a transcription factor.

Western blot analysis of TBX5 using anti-TBX5 antibody. Human whole cell lysates from HeLa, HepG2, MCF-7, and U2OS cells were resolved by 10% SDS-PAGE under reducing conditions and transferred to a nitrocellulose membrane. TBX5 was detected as a prominent band migrating at approximately 65-70 kDa across all tested cell lines. The predicted molecular weight of TBX5 based on amino acid sequence is approximately 58 kDa; the higher apparent molecular weight observed on SDS-PAGE is consistent with reported behavior of T-box transcription factors and may reflect post-translational modifications, altered SDS binding, or conformational properties that affect electrophoretic mobility rather than a difference in primary sequence.

TBX5 antibody targets T-box protein 5, a transcription factor encoded by the TBX5 gene that plays a central role in embryonic development and organ specification.
