Description

NCOA1 Antibody / Nuclear Receptor Coactivator 1 Antibody recognizes NCOA1, a transcriptional coregulator that functions as a coactivator for members of the nuclear receptor superfamily. NCOA1 is also widely known as SRC-1, or Steroid Receptor Coactivator 1, and is a founding member of the p160 nuclear receptor coactivator family. Following ligand-dependent activation of nuclear receptors, NCOA1 can associate with these transcription factors and facilitate recruitment of additional regulatory proteins to hormone-responsive genes. Through these interactions, NCOA1 helps connect receptor activation with chromatin regulation and transcriptional responses.

NCOA1 contains multiple LXXLL motifs that mediate interactions with ligand-bound nuclear receptors. The protein also contains transcriptional activation domains capable of recruiting chromatin-modifying factors such as CBP and p300. These interactions promote histone acetylation and changes in chromatin accessibility that facilitate transcription. NCOA1 itself has also been associated with histone acetyltransferase activity and is designated KAT13A, reflecting the close relationship between nuclear receptor coactivation and chromatin-dependent regulation of gene expression.

Nuclear Receptor Coactivator 1 participates in transcriptional responses mediated by estrogen, progesterone, androgen, glucocorticoid, thyroid hormone and other nuclear receptors. Its influence on transcription can depend on receptor type, ligand availability, cellular context and the abundance of other transcriptional coregulators. An NCOA1 Antibody can therefore support investigation of the molecular mechanisms that determine the magnitude and specificity of nuclear receptor-dependent gene expression and cellular responses to hormonal signals.

NCOA1 has important roles in endocrine and metabolic biology and has been investigated in reproductive tissues, hormone-responsive organs and metabolic regulation. Altered nuclear receptor coactivator expression or activity can modify hormone-dependent transcriptional programs and has also been studied in cancer biology. In particular, NCOA1/SRC-1 has attracted interest in hormone-responsive breast and prostate cancers, where changes in steroid receptor signaling and transcriptional coregulation may influence tumor progression and therapeutic response.

As a transcriptional coactivator positioned between activated nuclear receptors and the transcription machinery, NCOA1 provides a useful target for investigating hormone signaling, chromatin regulation and receptor-dependent gene expression. Its interactions with nuclear receptors and other transcriptional regulators also connect NCOA1 with broader mechanisms of cellular signal transduction.
