SNAP25 Antibody / Synaptic Vesicle Fusion Protein recognizes SNAP25, a 25 kDa protein and essential component of the neuronal SNARE machinery that regulates neurotransmitter release. SNAP25, also called synaptosomal-associated protein 25, is a membrane-associated t-SNARE that interacts with syntaxin and synaptobrevin/VAMP proteins to form the SNARE complex responsible for bringing synaptic vesicle and presynaptic plasma membranes together during exocytosis. Through this activity, SNAP25 has a central role in synaptic vesicle docking, priming and membrane fusion.

SNAP25 is highly associated with neurons and presynaptic terminals and is widely studied as a marker of synaptic and neuronal integrity. Its involvement in regulated exocytosis makes the protein important for studies of neurotransmission, synaptic plasticity and neurological disease. Two major neuronal isoforms, SNAP25a and SNAP25b, are generated through alternative splicing and show developmental and functional differences. SNAP25 also interacts with voltage-gated calcium channels and other proteins involved in controlling neurotransmitter secretion. Outside the nervous system, SNAP25 participates in regulated secretion in several neuroendocrine cell types, including pancreatic beta cells.

The importance of SNAP25 to membrane fusion is also demonstrated by its susceptibility to clostridial neurotoxins. SNAP25 is a substrate of botulinum neurotoxins, and proteolytic cleavage disrupts SNARE-mediated membrane fusion and transmitter secretion. Consequently, SNAP25 biology is relevant to research involving synaptic vesicle cycling, neurosecretion, presynaptic function and mechanisms of neurotoxin action.

SNAP25 is highly conserved among commonly studied mammalian species, supporting its use as a research target across a broad range of neuroscience models.
