SMAD3 / MADH3 Antibody Human Liver Cancer IHC. Immunohistochemistry was performed on paraffin-embedded human liver cancer tissue using SMAD3 / MADH3 Antibody at 2 ug/ml. Heat-induced epitope retrieval was carried out in pH 8 EDTA buffer, and the tissue was incubated with the primary antibody overnight at 4°C. Bound antibody was detected using an HRP-conjugated goat anti-rabbit IgG secondary antibody and DAB chromogen. Strong cytoplasmic staining is observed throughout the liver carcinoma cells, while the surrounding stromal tissue exhibits comparatively low background staining. These results demonstrate that this MADH3 Antibody is suitable for immunohistochemical detection of SMAD3 in formalin-fixed, paraffin-embedded human liver cancer tissue.
SMAD3 / MADH3 Antibody A549 Cell IF. Immunofluorescence was performed on human A549 cells using SMAD3 / MADH3 Antibody at 5 ug/ml together with an anti-Tubulin Alpha antibody. Cells underwent enzyme antigen retrieval for 15 minutes prior to incubation with the primary antibodies overnight at 4°C. SMAD3 was detected with an FITC-conjugated goat anti-rabbit IgG secondary antibody (green), while Tubulin Alpha was detected with a Cy3-conjugated goat anti-mouse IgG secondary antibody (red). The merged image demonstrates prominent nuclear localization of SMAD3 against the cytoplasmic microtubule network, confirming that this MADH3 Antibody is suitable for immunofluorescent detection of SMAD3 in cultured human cells.
SMAD3 / MADH3 Antibody Human, Rat and Mouse WB. Western blot analysis was performed using SMAD3 / MADH3 Antibody at 0.5 ug/ml. Lane 1: human A549 whole cell lysate; lane 2: human RT4 whole cell lysate; lane 3: human A431 whole cell lysate; lane 4: human PC-3 whole cell lysate; lane 5: rat brain tissue lysate; lane 6: rat ovary tissue lysate; lane 7: mouse brain tissue lysate. Proteins were separated on a 10% SDS-PAGE gel, transferred to a nitrocellulose membrane, and the blot was incubated with the primary antibody overnight at 4°C. Bound antibody was detected using an HRP-conjugated goat anti-rabbit IgG secondary antibody and enhanced chemiluminescence. A specific immunoreactive band is detected at approximately 50-55 kDa in all tested samples, consistent with the expected molecular weight of SMAD3 (~48 kDa). These results demonstrate that this MADH3 Antibody is suitable for Western blot detection of SMAD3 in human, rat, and mouse samples.
