SDHB Antibody Protein Microarray Specificity Validation. HuProt protein microarray analysis was performed with SDHB Antibody, clone SDHB/2382, against more than 19,000 full-length human proteins. SDHB ranked as the top target with a Z-score of 118.67 and an S-score of 114.74, showing exceptional separation from the next-ranked proteins. The Z-score represents signal strength in standard deviations above the array mean, while the S-score measures the difference between successive ranked Z-scores and therefore reflects relative target specificity. These results demonstrate highly specific recognition of SDHB by clone SDHB/2382 in this protein microarray assay.

SDHB Antibody / Complex II Monoclonal Antibody detects succinate dehydrogenase complex iron sulfur subunit B, a nuclear-encoded component of mitochondrial respiratory Complex II. SDHB functions together with SDHA, SDHC and SDHD to form succinate dehydrogenase, an enzyme complex located in the inner mitochondrial membrane. Succinate dehydrogenase is unusual among respiratory chain complexes because it participates directly in both the tricarboxylic acid (TCA) cycle and mitochondrial electron transport, linking intermediary metabolism with oxidative phosphorylation.

During the TCA cycle, succinate dehydrogenase catalyzes the oxidation of succinate to fumarate. Electrons generated during this reaction are initially accepted by flavin adenine dinucleotide associated with SDHA and subsequently transferred through a series of iron-sulfur clusters contained within SDHB. Electron transfer then proceeds toward ubiquinone through the membrane-associated SDHC and SDHD components. SDHB therefore occupies a central position in the electron-transfer activity of mitochondrial Complex II.

Proper SDHB expression and Complex II assembly are important for mitochondrial respiration and cellular metabolic homeostasis. Loss or dysfunction of an SDH component can reduce succinate dehydrogenase activity and cause intracellular accumulation of succinate. Elevated succinate can inhibit alpha-ketoglutarate-dependent enzymes and alter oxygen-sensing and epigenetic regulatory mechanisms. These effects connect mitochondrial metabolic dysfunction with broader changes in cell signaling and gene regulation.

SDHB is particularly important in cancer research because pathogenic alterations of the SDHB gene are associated with hereditary paraganglioma and pheochromocytoma syndromes and have also been investigated in other SDH-deficient tumors. Succinate accumulation resulting from impaired SDH activity can contribute to tumor-associated metabolic reprogramming, leading to the classification of succinate as an oncometabolite. Evaluation of SDHB protein expression is consequently useful in research examining mitochondrial dysfunction, tumor metabolism and the molecular consequences of succinate dehydrogenase deficiency.

Clone SDHB/2382 is a mouse monoclonal antibody developed for recognition of SDHB. Protein microarray validation provides an additional approach for assessing antibody target recognition across a large collection of human proteins, complementing conventional experimental validation such as Western blot analysis. The defined monoclonal nature of SDHB/2382 also provides consistent recognition of its target epitope for studies of SDHB protein expression.
