S100 Calcium-Binding Protein B antibody, also known as Pan-Tissue Validated Marker antibody, recognizes S100 calcium-binding protein B, a small EF-hand calcium sensor encoded by the human S100B gene located on chromosome 21q22.3. S100 calcium-binding protein B is a member of the S100 family of calcium-binding proteins that function as intracellular regulators of cytoskeletal dynamics, enzyme activity, and transcriptional control. The protein forms homodimers and undergoes conformational change upon calcium binding, enabling interaction with diverse signaling partners.
S100B is most prominently expressed in astrocytes within the central nervous system, where it contributes to calcium-dependent signaling and regulation of glial cell morphology. However, expression is not restricted to neural tissue. S100 calcium-binding protein B is also detected in melanocytes, Schwann cells, adipocytes, chondrocytes, and selected epithelial and stromal cell populations. Because of this lineage-specific but multi-tissue distribution, S100B has become an important marker in both neurobiology and oncology research.

1. Optimal dilution of the S100 Calcium-Binding Protein B antibody should be determined by the researcher.
2. This S100 Calcium-Binding Protein B antibody is recombinantly produced by expression in human HEK293 cells.
3. Manual Protocol: Freshly cut sections should be used (less than 10 days between cutting and staining). Heat-induced antigen retrieval for 5 minutes in an autoclave at 121oC in pH 7.8 Target Retrieval Solution buffer. Apply the antibody at a dilution of 1:150 at 37oC for 60 minutes. Visualization of bound antibody by the EnVision Kit (Dako, Agilent) according to the manufacturer's directions.
