Renal Lineage Marker PAX8 antibody recognizes PAX8 protein, a nuclear transcription factor encoded by the PAX8 gene and a critical regulator of renal epithelial differentiation. PAX8 protein is a member of the paired box family of transcription factors and plays a central role in kidney development and epithelial lineage specification. Because of its consistent expression in renal tubular epithelial cells, PAX8 antibody is widely used in research focused on renal biology and renal-derived tumors.
PAX8 contains a conserved paired DNA-binding domain that enables regulation of genes involved in organogenesis and epithelial differentiation. During embryonic kidney development, PAX8 contributes to nephric lineage commitment and maturation of renal epithelial structures. In adult tissues, PAX8 protein remains expressed in renal tubular epithelial cells, where it maintains aspects of epithelial identity. Detection of PAX8 protein therefore reflects renal epithelial lineage and differentiation status.
PAX8 protein localizes to the nucleus, consistent with its role as a transcription factor. Beyond the kidney, PAX8 expression is also observed in thyroid follicular cells and Mullerian-derived epithelium, including fallopian tube and endometrium. In pathologic settings, PAX8 expression is commonly retained in renal cell carcinomas, including clear cell, papillary, and chromophobe subtypes. Because of this expression pattern, PAX8 antibody is frequently used in studies evaluating tumor origin and distinguishing renal carcinomas from other malignancies.
The PAX8 gene is located on chromosome 2q14 and produces multiple isoforms through alternative splicing. Dysregulated expression and gene rearrangements involving PAX8 have been reported in thyroid and other epithelial tumors. Renal Lineage Marker PAX8 antibody clone MSVA-708R is a recombinant rabbit monoclonal antibody developed to detect PAX8 protein in research applications. As a recombinant monoclonal antibody, clone MSVA-708R supports consistent nuclear detection of PAX8 protein in renal epithelial tissues and related tumor models.
1. Optimal dilution of the Renal Lineage Marker PAX8/PAX8 Protein antibody should be determined by the researcher.
2. This Renal Lineage Marker PAX8/PAX8 Protein antibody is recombinantly produced by expression in human HEK293 cells.
3. Manual Protocol: Freshly cut sections should be used (less than 10 days between cutting and staining). Heat-induced antigen retrieval for 5 minutes in an autoclave at 121oC in pH 7.8 Target Retrieval Solution buffer. Apply the antibody at a dilution of 1:150 at 37oC for 60 minutes. Visualization of bound antibody by the EnVision Kit (Dako, Agilent) according to the manufacturer's directions.
A recombinant fragment (around amino acids 150-300) of human PAX8 (exact sequence is proprietary) was used as the immunogen for the Renal Lineage Marker PAX8/PAX8 Protein antibody.
