Survivin Antibody BIRC5/8936R / Apoptosis Inhibitor Survivin Antibody recognizes survivin, a member of the inhibitor of apoptosis protein (IAP) family encoded by the BIRC5 gene. Survivin is a small regulatory protein with important functions in both cell survival and mitosis. Unlike many other IAP family members, survivin contains a single baculoviral IAP repeat domain and lacks the typical C-terminal RING finger domain. Its dual roles in suppressing apoptosis and regulating cell division have made BIRC5 an extensively studied protein in cancer and cell biology.

Survivin participates in the chromosomal passenger complex together with Aurora B kinase, INCENP, and borealin/CDCA8. This complex regulates several critical events during mitosis, including chromosome alignment and segregation, correction of kinetochore-microtubule attachments, spindle checkpoint function, and cytokinesis. Survivin contributes to localization and activity of the complex as cells progress through mitosis, linking BIRC5 expression closely with cellular proliferation.

BIRC5 is also associated with resistance to programmed cell death. Through interactions with components of apoptotic signaling pathways, survivin can promote cell survival and reduce susceptibility to apoptosis. The balance between its mitotic and anti-apoptotic functions is influenced by protein localization, molecular interactions, and cell-cycle status, allowing survivin to coordinate mechanisms governing cell division and survival.

Survivin expression is generally low in most differentiated adult tissues but is elevated during development and in many malignancies. Increased BIRC5 expression has been investigated in numerous solid tumors and hematologic cancers and has been associated with tumor-cell proliferation, resistance to apoptosis, disease progression, and treatment response. These characteristics have made survivin an important subject in cancer biomarker and therapeutic-target research.

Alternative splicing of BIRC5 produces several survivin isoforms, including survivin-2B and survivin-deltaEx3, which may differ in localization and biological activity. Studies of BIRC5 therefore encompass apoptosis regulation, mitotic control, cell proliferation, cancer biology, and the functional significance of survivin isoforms.
