Succinate Dehydrogenase B Antibody Human HepG2 WB. Western blot testing of human HepG2 cell lysate was performed with Succinate Dehydrogenase B Antibody. A strong specific band is detected at approximately 29 kDa, within the predicted molecular weight range of 25-32 kDa for SDHB. Succinate Dehydrogenase B Antibody Human Hepatocellular Carcinoma IHC. IHC staining of FFPE human hepatocellular carcinoma tissue was performed with Succinate Dehydrogenase B Antibody, clone SDHB/8974R. Heat-induced epitope retrieval was performed by boiling tissue sections in pH 9.0, 10 mM Tris with 1 mM EDTA for 20 minutes, followed by cooling prior to staining. Strong cytoplasmic SDHB staining is observed throughout the tumor cells. Succinate Dehydrogenase B Antibody / SDHB Recombinant Antibody detects SDHB, the iron-sulfur subunit of mitochondrial succinate dehydrogenase, also known as respiratory Complex II. SDHB is encoded by the nuclear SDHB gene and functions together with SDHA, SDHC and SDHD to form the succinate dehydrogenase complex within the inner mitochondrial membrane. Complex II has a distinctive metabolic role because it participates in both the tricarboxylic acid (TCA) cycle and the mitochondrial electron transport chain.

During the TCA cycle, succinate dehydrogenase catalyzes the oxidation of succinate to fumarate. Electrons generated during this reaction are transferred from the flavin-containing SDHA subunit through a series of iron-sulfur clusters located within SDHB. They are subsequently transferred toward ubiquinone through the membrane-associated components of the complex. This process connects TCA cycle metabolism with mitochondrial respiration and contributes to efficient cellular energy production.

SDHB is particularly important for the electron-transfer function of Complex II. Disruption of SDHB or other succinate dehydrogenase components can impair mitochondrial respiration and result in intracellular accumulation of succinate. Elevated succinate can influence cellular signaling and gene regulation through inhibition of alpha-ketoglutarate-dependent enzymes, linking mitochondrial metabolic dysfunction with broader changes in cellular physiology.

Alterations affecting SDHB are extensively studied in cancer biology. Germline SDHB mutations are associated with hereditary paraganglioma and pheochromocytoma syndromes, and disruption of succinate dehydrogenase function has also been investigated in other tumor types. Succinate accumulation resulting from SDH deficiency can contribute to altered oxygen-sensing and epigenetic pathways, supporting the characterization of succinate as an oncometabolite in SDH-deficient disease.
