NF-H Antibody / NF200 Antibody detects neurofilament heavy chain (NF-H), a neuronal intermediate filament protein encoded by the NEFH gene. Also commonly referred to as neurofilament 200 or NF200, NF-H is a major structural component of the neuronal cytoskeleton. Neurofilaments are particularly abundant within axons, where they contribute to mechanical stability, organization of neuronal processes and regulation of axonal caliber.
Neurofilaments are composed primarily of light, medium and heavy subunits, commonly designated NF-L, NF-M and NF-H. Neurofilament heavy chain has an apparent molecular weight of approximately 200 kDa and contains an extended C-terminal tail rich in repeated lysine-serine-proline motifs. Extensive phosphorylation of this region influences neurofilament organization and interactions with other cytoskeletal components, helping establish the specialized architecture of mature axons.
NF200 Ab is widely used in neuroscience research to investigate neuronal structures and axonal organization. NF-H expression is particularly useful for visualizing neuronal processes and examining neuronal differentiation, maturation and nervous system architecture. Neurofilament proteins are also studied in peripheral nerves and neural or neuroendocrine tissues, providing useful markers for research involving neuronal lineage and differentiation.
Changes in NF-H expression, organization and phosphorylation can accompany neuronal injury and neurodegeneration. Disruption of normal neurofilament transport may contribute to abnormal protein accumulation and altered axonal structure. NF-H is therefore investigated in studies of neuroaxonal damage and neurological disorders, including amyotrophic lateral sclerosis, multiple sclerosis and other conditions involving neuronal or axonal degeneration.
Clone NFHP-1R is a recombinant rabbit monoclonal antibody developed against recombinant human neurofilament heavy chain. The antibody has been evaluated by Western blot and immunohistochemistry for detection of human NF-H. Recombinant production provides a defined antibody sequence and supports consistent performance across manufacturing lots. The stated application concentrations are suggested starting points. Titration of the NF-H Antibody / NF200 Antibody may be required due to differences in protocols and secondary/substrate sensitivity.
1. The prediluted format is supplied in a dropper bottle and is optimized for use in IHC. After epitope retrieval step (if required), drip mAb solution onto the tissue section and incubate at RT for 30 min.
Recombinant human protein (heavy subunit) was used as the immunogen for the recombinant NF-H antibody.
Store the recombinant NF-H antibody at 2-8oC (with azide) or aliquot and store at -20oC or colder (without azide).
